Leaves were one of the vital organs of plants. The morphological and anatomical indicators of leaves were observed frequently in the process of teaching and research work. Research about observation methods of morphologica and anatomical indicators more confined to leaves’ epidermal cells, and there were no detail researches for man y factors which influence observed effects. The fewer methods were used for observing leaves’ transection and nervation, and ha d short-coming of complex operation sequence, time-consuming, poor slicing, etc. In order to solve the above problems, corn of monocot grramineae herb plants and Lonicera edulis of woody Dicotyl edonous were as material by the method of orthogonal test to detailed systematic research about observation methods of epidermis and transection and nervation in two different kinds of plant leaves, got simple and quick observation methods of their morphological and anatomical indicators with condition. They were used for measuring many quantitative indicators of epidermis, mesophyll and nervation in two kinds of plant leaves. The results provided technical support for quickly and easily observing morphological and anatomical indicators of plan leaves with research the mechanism of leaf growth. The main conclusions are as follows:(1) Nail polish blotting and gelatin blotting were simple qu ick observation methods of epidermis and transection in maize leaves, segregation method of Na Cl O maceration and peroxyacetic acid maceration were the best observation methods of nervation.Among, the optimal conditions about observing the indicators of maize leaves’ epidermis by nail polish blotting contained 12 minutes of nail polish solidification time, 1.5×1.5 cm2 of square of adhesive tape with 60° of quickly removing the adhesive tape, by gelatin blotting contained 21% concentration of gelatin, 8 minutes clotting time of the gelatin, 0.018g·100m L-1 dosage of methyl green, 22°C ambient temperature and 11 seconds standing time of the leaves on the gelatin; The optimal conditions about observing the indicators of maize leaves’ transection by nail polish blotting contained fixed six leaves and 50 seconds of nail polish solidification time and 60° of slowly removing the adhesive tape or fixed six leaves and 60 seconds of nail polish solidification time and 30° of slowly removing the adhesive tape, by gelatin blotting contained 8 minutes clotting time of the gelatin, 18% concentration of gelatin, 0.045g·100m L-1 dosage of methyl green, 22°C ambient temperature and 5 seconds standing time of the leaves on the gelatin.The optimal conditions about observing the indicators of maize leaves’ nervation by segregation method of Na ClO maceration contained boiling for 4 min, then 10% Na OH 60°C water bath for 30 min, then 4% Na ClO 20°C water bath for 30 min, by peroxyacetic acid maceration contained 14% peroxyacetic acid 70°C water bath for 3h.(2) Nail polish blotting and gelatin blotting were simple quick observation methods of epidermis and transection in Lonicera edulis leaves.The optimal conditions about observing the indicators of maize transection leaves by gelatin blotting were the same as maize leaves’. segregation method of Na ClO maceration and peroxyacetic acid maceration were the best observation methods of nervation.Among, the optimal conditions about observing the indicators of Lonicera edulis leaves’ epidermis by nail polish blotting contained 20 minutes of nail polish solidification time, 1×1 cm2 of square of adhesive tape and 45° or 60°of quickly removing the adhesive tapeg, by gelatin blotting contained 24% concentration of gelatin,8 minutes clotting time of the gelatin, 0.036 g ·100mL-1 dosage of methyl green, 20°C ambient temperature and 6 seconds standing time of the leaves on the gelatin; The optimal conditions about observing the indicators of Lonicera edulis leaves’ transection by nail polish blotting contained fixed five leaves and 70 seconds of nail polish solidification time and 60° of slowly removing the adhesive tape.The optimal conditions about observing the indicators of Lonicera edulis leaves’ nervation by segregation method of Na Cl O maceration contained boiling for 5 min, then 8% NaOH 60°C water bath for 1.5h, then 4% NaCl O 40°C water bath for 35 min, by peroxyacetic acid maceration contained 14% peroxyacetic acid 60°C water bath for 3.5 h or 12% peroxyacetic acid 70°C water bath for 3.5 h.
