objective: to investigate inhibition and antioxidant activities of the different extractions of xinjiang Rubus idaeus L. on protein tyrosine phosphatase 1 b(PTP1B),exploring effective parts with antioxidant and PTPIB inhibitory activities. Studied the cotent change of total polyphenols and total flavonoids of Rubus idaeus L. leaf in a year, to explore the change rule and antioxidant activity. Investigated the effection of different factors on the flavonoids extraction yield and antioxidant activity of Rubus idaeus L. stem, determined the best extraction technology of flavonoids of Rubus idaeus L. stem. using accurate mass measurement and characteristic fragmentation patterns finded 16 flavonoids, including 4 quercetin derivatives, 2 luteolin derivatives, 8 kaempferol derivatives and 2 isorhamnetin derivatives. Methed:The different polar fractions of Rubus idaeus L. were extracted by ethanol and fractional extraction method. Then the total flavonoids content of different polar fractions were determined. The anthocyanin fractions of fruit were purified by AB-8 macroporous resin; Their PTP1 B inhibition were measuresd by enzymology method, and antioxidant activities were measuresd by ABTS, DPPH method. Determined the total flavonoids content and antioxidant activities of ethanol extracts of the different periods of Rubus idaeus L. leaf. the influence of extraction parameters(different solvents, ethanol concentration, temperature, time, liquid to solid ratio) on flavonoids extraction rate and antioxidant activity of Rubus idaeus L. stem were determined. employed High performance liquid chromatography coupled with electrospray ionization quadrupole- time- of-flight tandem mass spectrometry identificated flavonoids of Rubus idaeus L. Leaf. Result: Followed by AB-8 macroporous resin purification, anthocyanin contents of Rubus idaeus L. fruit extract increased from 12.7±0.14 mg·g-1 to 238.1±1.18 mg·g-1, its antioxidant activity and PTP1 B inhibitory activity were also significantly increased. The ethyl acetate fraction of Rubus idaeus L.leaf had the highest total flavonoids content(456.86±0.70mg·g-1); The ethyl acetate fraction of Rubus idaeus L.leaf had the highest antioxidant activity, and its ABTS and DPPH radical scavenging capacity were 3.22±0.02 and 2.95±0.01 mmol Trolox·g-1 equivalent, repectively; The anthocyanin fractions of Rubus idaeus L. fruit had the highest PTP1 B inhibitory activity(IC50=0.22±0.04 μg/mL). the content of total polyphenols and total flavonoids of Rubus idaeus L. leaves had a overall upward trend in a year. the minimum value of the content of total flavonoids(2.116 mg/g) and total polyphenols(38.279 mg/g) were in may all, the maximum value of the content of total flavonoids(2.934 mg/g) and total polyphenols(53.491 mg/g) were in October all. There were a certain correlation among the total flavonoids, total polyphenol and the scavenging rate of DPPH. The optimal flavonoids extraction conditions(80% ethanol, 70℃, 45 min, solid-liquid ratio of 1:40) were obtained. studied flavonoids purification conditions of the Rubus idaeus L. stems, found that AB-8 macroporous resin purification effection is best, content of total flavonoids significantly increased after purification. among identificated 16 flavonoids of Rubus idaeus L.leaf, Compounds 3 and 4, compounds 6 and 7, compounds 15 and 16, were isomers with different aglycones and different saccharides. Compounds 8, 9 and 10 were isomers with same aglycone and same saccharide but different substituent positions.compounds11and12 wereisomerswithsameaglyconebutdifferentsaccharides.compounds2,8,9and10 possessedthesamesubstituentsaccharideofglycuronicacid.conclusion:thedifferentfractionsofrubusidaeusl.havehighflavonoidcontentsandgoodantioxidantactivityandptp1 binhibition.then-butylalcoholfractionofrubusidaeusl.leafwasthemainactivefraction,anthocyaninofrubusidaeusl.fruitwasthemainactiveingredient.thecontentofflavonoids,polyphenolandthescavengingrateofdpphoftherubusidaeusl.leafwasthehighestinoctober,sotheoptimaltimeofcollectingraspberryleavesisoctober.rubusidaeusl.stemhashighflavonoidscontentandantioxidantactivity,anditispotentialresourcesofnaturalantioxidants.identificatedtheactiveingredientisolatedfromrubusidaeusl.leaf,foundthatmostofthemarefirstisolatedfromrubusidaeusl.leaf,thediscoveryofthesecompoundstofindingofplantmedicineofrubusidaeusl.leafarevaluable.
