Rian is an identified maternal expressed long non-coding RNA(lnc RNA), which is located in mouse distal chromosome 12. And it contains three of long transcripts Irm,Meg8 and AB063319. A large study showed that Rian plays an important role in controlling biological processes. On the basis of others’ research achievements, as the main transcript of Rian, Irm is frequently expression in developing skeletal muscle and the expression is rising. But in the mature skeletal muscle Irm is low expression,which is in agreement with many genes, which are regulated to development of skeletal muscle, suggesting Irm plays an important role in skeletal muscle development.Taking the C2C12 and primary muscle cells as the study subject, we first examined the expression pattern of Irm during the proliferation and differentiation of the C2C12 and primary muscle cells in vitro. Secondly by means of overexpressing and down-regulating the expression of Irm, we study the influence of Irm to proliferation,migration and differentiation of the skeletal muscle cells. To further investigate the function of Irm to proliferation and differentiation, we use the method of whole genome gene expression profiling sequencing to screen the differential expression genes associated with skeletal muscle development in the end.Morphological observation and analysis of the results show that up-regulating Irm expression will suppress cell proliferation and migration, but it will accelerate cell differentiation and up-regulated Myo D and Myogenin expression. Nevertheless,down-regulating Irm expression will accelerate cell proliferation and migration, but it will suppress cell differentiation and down-regulated Myo D and Myogenin expression.According to the microarray data analysis and the results of q RT-PCR, changing the Irm expression have effect on the upstream and downstream genes expression of the p38 MAPK.These results indicated that expression level of Irm inhibited skeletal muscle cell proliferation and migration, and accelerate skeletal muscle cell differentiation. we suggest that the it maybe do that by the p38 MAPK signal pathways.The results of the study will supply the extent theory foundation for well understanding the significance of lnc RNA to disease, development and evolution.
