Oil distribute widely in plants,animals and algae,et al.lt contains a variety of long-chain polyunsaturated fatty acids which have biologically actives.They have many special functions such as reducing blood fat content,reducing triglyceride and cholesterol levels content,preventing cardiovascular disease,protecting eyesight and improving intelligence.At present,many oil have been extracted from many marine organisms,such as deep sea fish oil and Antarctic krill.Although Krill,Euphausia pacifica have rich in storage capacity in the nature, there are still a lot of resources have not been exploited.The present research on Euphausia pacifica is mainly focus on its life history, multiplication, developmental,migration in day and night, vertical distribution and special enzymes and ecology,but its nutrition and oil conponents are less reported. Meanwhile, there are no report on the pharmacokinetics of Euphausia pacifica oil, and the changes of the concentration of fatty acids in blood after taking have not been reported and need more research.In this paper,we tried to evaluate the technology of oil extraction from Euphausia pacifica and analysed its components. In addition,we also researched the pharmacokinetics charateristic of Euphausia pacifica oil in rabbits,those results will provide theoretical basis for the development and application of scientific evidence of Euphausia pacifica oil in the furture.Research results:(1)The extraction process optimization of Euphausia pacificaThe optimal conditions of extracting oil from Euphausia pacifica were investigated using organic solvent. The results showed that 95% ethanol was the best extraction solvent. And the yield was 23.48±0.10%, when it was extracted at 60℃ for 6h, powder of Euphausia pacifica: complex of organic solvent ratio was 1:10 (w:v)(2)Fat composition analysis of Euphausia Pacifica oilNational standards were used to comprehensively detect and evaluate the oil of Euphausia Pacifica.The content of water in the dry powder of Euphausia Pacifica was 0.14±0.02%;The content of phospholipids in the oil was 27.74±0.13%;The content of peroxide value in the oil was 4.73±0.04 mmol·kg-1;The content of acid value in the oil was 7.08±0.15(KOH,mg·g-1);The content of saponification value in the oil was 145.44±3.75(KOH,mg·g-1);The content of fluorine in the oil was 5.60±0.75mg·kg-1;The content of astaxanthin in the oil was 200±6mg/100g.(3)The fatty acid composition of Euphausia pacifica oil.The oil was mainly composed of twelve kinds of fatty acids which analyzed by gas chromatography. Four saturated fatty acid were identified from the oil. The total yield of saturated fatty acid was as high as 26.00%. Eight unsaturated fatty acid were identified from the oil.The total yield of unsaturated fatty acids was as high as 55.90%, The content of monouns unsaturated fatty acids was 20.00%, poly unsaturated fatty acids was 35.9%,the content of EPA and DHA was 16.7% and 15.2%,respectively.(4)To investigate the pharmacokinetic characteristics of Euphausia pacifica oilThe plasma concentration of fish oil,Antarctic krill oil and Euphausia pacifica oil in rabbits was measured using GC-MS method and the pharmacokinetic parameters were calculated by the 3P97 program.The pharmacokinetic results showed that there was no significant difference in bioavailability for the three kinds of oil using determination of EPA as indictor;the tmax of two kinds of krill oil was longer than fish oil;the peak plasma concentration of Euphausia pacifica oil was higher than the other two kinds of oil;Using determination of DHA as indictor,the bioavailability of Euphausia superba oil was much higher than Antarctic krill;the tmax of Antarctic krill oil was longer than the other two kinds of oil;the peak plasma concentration of fish oil was higher than the other two kinds of krill oil.This study conducted a effectively extraction process of oil from Euphausia pacifica and analysed the different components of the oil,established the foundation of Euphausia pacifica oil for further research and the market application;The results of vivo pharmacokinetic process in rabbit about the EPA,DHA of fish oil,Antarctic krill oil and Euphausia pacifica oil made foundation to illuminate the theory of oil absorption both in vivo and clinical medicine application.
