Xanthoceras sorbifolia Bunge is a unique and rare woody oil tree species in China, and is widely planted in the three northern areas of China, plant oil is one of the three major nutrients of humans. It becomes a goal for many studies to regulate fatty acid content and composition by using gene engineering. According to research reports, the fatty acid elongase 1 (FAE1) and fatty acid desaturase 2 (FAD2) are key enzymes of long-chain fatty acids and polyunsaturated fatty acid synthesis, and FAE1/FAD2 gene control the fatty acid composition of plant oil.In this study, primers were designed based on plants FAE1, FAD2 squences recorded in the NCBI, and full-length FAE1, FAD2 cDNA (named As XsFAEl and XsFAD2-2) was cloned by RT-PCR and RACE from seeds of Xanthoceras sorbifolia Bunge. XsFAEl has a 1914 bp full-length cDNA containing a 1491 bp ORF, which encode 497 amino acids. XsFAD2-2 has a 1605 bp full-length cDNA, containing a 1152 bp ORF, which encode 383 amino acids. Based on this, the expression patterns of two genes were also analyzed by quantitative PCR. The results show that XsFAE1 and XsFAD2-2 genes were expressed mainly in the seed, in witch XsFAE1 gene expression was highest at seed 40 days after flowering, and XsFAD2-2 gene expression was highest seed at 80 days after flowering.This study successfully obtained full-length cDNA of XsFAE1 and XsFAD2-2 gene. The above results may lay the foundation of expression of XsFAE1 and XsFAD2 genes in the oil crop seeds. |
