Objective Through observing the expression level of PRMT5 and β- Catenin in different development period of mouse kidney, to explore the influence of PRMT5 and β-Catenin in the development of mouse kidney, and provide experimental basis for further discussion of the early renal unit.Methods 1. Immunohistochemical PV method was used to detect PRMT5 and β-Catenin in different embryo age(E) of 14, 16, 18 D of fetal and postnatal(N) expression in 1, 7, 14,21 and 42 d in rat kidney tissues, further observation and analysis that the correlation and expression in renaltissues of PRMT5 and β-Catenin in mouse embryos and postnatal kidney tissues. 2. RT-PCR Technology was used to detect that the expression of m RNA about PRMT5 and β-Catenin in different embryonic age(E) of 14, 16 and 18 d in fetal and different day ages postnatal(N) of 1, 7, 14, 21 and 42 d rats kidney tissue samples. 3. Western Blot Technology was used to detect that the expression of PRMT5 and β-Catenin in different embryonic age(E) of 14,16 and 18 d in fetal and different day ages postnatal(N) of 1,7,14,21 and 42 d rats kidney tissue samples,analyze the relationship in each group and the changes of gene expression in PRMT5 and β-Catenin.Results 1. PRMT5, β-Catenin gene expression at the start of the ureteric bud fromthe E14 d. Then with the kidney gradually mature, β-Catenin was mainly expressed in the distal tubule and collecting duct, in the proximal tubule rare expression, no expression in renal corpuscle;PRMT5 was mainly expressed in the distal tubule, proximal tubules and glomeruli in rare expression. 2. RT-PCR and Western Blot results showed: in different stages of renal tissue of mice with kidney development, from E14 d to N1 d, PRMT5 and β-Catenin expression increased gradually in the kidney, and from N1 d to N42 d expression gradually decreased. 3. In different stages of mouse kidney development, the trend of expression about β-Catenin and PRMT5 in renal tissue is basically the same, showed a significant positive correlation(r=0.806,P<0.001).Conclusions In different stages of mouse kidney development, PRMT5 and β-Catenin was mainly expressed in the distal tubule, rare expression in the proximal tubule. Correlation analysis showed that trend of expression about β-Catenin and PRMT5 in renal development in different stages is basically the same, the two had a significant positive correlation, indicating that PRMT5 may through the Wnt/β-Catenin signaling pathway to effect of mouse kidney development.
