Purpose:To find out the changes of the structure and weight in kidney tissues, we can do anexperiment on the activity of lactate dehydrogenase(LDH), Na+-K+ATP enzyme and theexpression of Caspase-3, Bax protein, the serum creatinine as well as the urea nitrogen levelsin blood after the the X-ray with different dosage. To know the comprehensive effects ofX-ray radiation with different doses on kidney tissues in mice, thus, we can analyze the cellinjury and apoptosis mechanisms under different X-ray radiation conditions.Method:96mice were divided into4groups by chance,24mice for every group, they weresystemic irradiated with different dosages (0,1,3,5Gy) of X-ray for three days in a row. For5,10,15and20d after irradiation, changes of kidney weight were examined. We can observedthe changes in the organizational structure of the kidney by Biological Microscopy, and examthe content of the serum creatinine and the urea nitrogen in blood. The activity of LDH,Na+-K+ATP enzyme on kidney tissue, by colorimetry, the Caspase-3and Bax protein levels onkidney tissue, by immunohistochemistry.Result:1) To some degree,the kidney weight increased at different rate relatively after thesystemic X-ray irradiation compared to the control group. Compared to the controlgroup,there was an increase in the kidney weight after1Gy radiation, but the increase was notsignificant?P>0.05). Compared to the control group, the kidney weight with3Gy and5Gyradiation increased significantly, the difference was significant or very significant?P?0.05orP?0.01). After5Gy radiation for5d, the increase in kidney weight was the most in contrastto other groups. Compare to the control group, the increase was0.11g. And the increase of thekidney weight is the least after1Gy(5,10,15,20)radiation, compare to the control group itincreased only0.02g.2) The serum creatinine levels in the experimental group compared with control groupincreased in varying degrees after the systemic X-ray irradiation. The serum creatinine levelswith1Gy radiation for5d and15d, the difference was very significant compared with thecontrol group?P<0.01?. The serum creatinine levels with3Gy radiation for5d,10d and20d, the difference was very significant compared with the control group?P<0.01?. After1Gy radiation for20d, the increase ofcreatinine level was the least, compare to the control group it increased only1.5?mol/L. After5Gy radiation for10d the increase of creatinine level was the largest with the other groups,compare to the control group it increased16.11?mol/L.3) Compared with control group,the urea nitrogen content in the experimental groupincreased at different rate after the systemic X-ray irradiation.With1Gy radiation for5d and15d,. the increase of urea nitrogen content was very significant in contrast to the control group?P<0.01?. And the urea nitrogen content with3Gy radiation for5d,10d and15d, thedifference was significant compared with the control group(P<0.05), the urea nitrogen contentwith5Gy radiation for10d, the difference was significant compared with the controlgroup(P<0.05). After1Gy radiation for20d, the increase of urea nitrogen content was theleast, for7.09mmol/L, compared to the control group, it increased only0.03mmol/L. After1Gy radiation for5d, the increase of urea nitrogen content was the most,9.00mmol/L.And itincreased1.92mmol/Lcompared the control group.4) Compared with control group, the activity of LDH increased at different rate after thesystemic X-ray irradiation. And as for the activity of LDH with1Gy radiation for5d,10d and15d, the difference was very significant?P<0.01?, the activity of LDH with3Gy radiation for10d,15d and20d, the difference was significant (P<0.05). As far as the activity of LDH with5Gy radiation for10d, the difference was significant (P<0.05). After1Gy radiation for20d,the increase of activity of LDH was the least, only3080?mol/min, compared to the controlgroup, it increased only960?mol/min. And after5Gy radiation for10d, the increase ofactivity of LDH was the most, only4910?mol/min, it increased2730?mol/min.5) Compared with control group the activity of Na+-K+ATP enzyme decreased atdifferent level after the systemic X-ray irradiation. The activity of Na+-K+ATP enzyme with1Gy radiation for5d and10d, the difference was very significant compared with the controlgroup?P<0.01?. The activity of Na+-K+ATP enzyme with3Gy radiation for10d,15d and20d,the difference was significant compared with the control group(P<0.05). The activity ofactivity of Na+-K+ATP enzyme with5Gy radiation for5d and10d, the difference wassignificant compared with the control group(P<0.05). After1Gy radiation for20d, the decrease in activity of Na+-K+ATP enzyme was the least, only11.28?molpi/mgprot/hour,compared to the control group it decreased only4.13?molpi/mgprot/hour. After5Gy radiationfor10d, the decrease of activity of Na+-K+ATP enzyme was the most,15.32?molpi/mgprot/hour, it increased11.08?molpi/mgprot/hour compared the control group.6) Immunohistochemistry showed that the expression of glomerular cells was notobvious, and there was no positive yellow in glomerular endothelial cells. However, there waspositive for yellow on the cellular cytoplasm and cell membrane of mouse renal tubular cells.The positive expression enhanced gradually with the increasing of radiation dose and time,which means the increase in the expression of Caspase-3protein compare to the controlgroup.7) Immunohistochemistry showed that the expression of Bax protein focused on parts ofthe cytoplasm and cell membrane of tubules and there were brown or yellow stained, but therewas no expression in glomerular endothelial cells, which means the expression of Bax proteinincreased at different rate after the X-ray radiation. The result shows the higher levels ofradiation,and the stronger expression of Bax protein.Conclusion:1) The kidney weight increased obviously in varying degrees.2) The X-ray radiation influence the organizational structure of mouse kidney. Thekidney tissues' structure injured at different rate. The largest the dose of radiation is, the mostobviously it damaged.3) The urea nitrogen content and the serum creatinine levels increased at different rateobviously after the systemic X-ray irradiation.4)After the X-ray radiation, kidney tissue lactate dehydrogenase levels increased, andproportional to the radiation dose.5)After the X-ray radiation, kidney tissue Na+-K+ATP enzyme activity decreased;compared with the control group, the difference was significant or very significant.6)After the X-ray radiation, kidney tissue apoptotic protein Bax protein, Caspase-3protein abundantly expressed.
