Water is an essential component during plant life cycle. The production loss caused by water deficit surpassed the total loss caused by other factors. After subjected to lrought stress, various physiological processes of most of the plants are affected in lifferent degree, so the cultivation of new resistant crop varieties has become an mportant appoach to cope with drought threaten.Caragana intermedia Kuang et H.C.Fu belongs to Caragana species, and widely istributed in Northwest China, is an important cultivated shrub with high ecological and prage value. Duting the long process of evolution, C. intermedia adapts to all kinds of armful growth environment, have the ability of to tolerate drought, cold, heat, aline-alkaline. The past studies on stress resistance ability of C. intermedia mainly >cused on the physiological, ecological and morphological aspects, the molecular lechanisms of stress resistance research has just started. In order to search for enes related to drought tolerance of C. intermedia, a full-length cDNA Library fleaves under drought stress was constructed.In this study, the 15-day soil drought in C. intermedia seedlings we chosen as sample. he full-length cDNAs were synthesized by the SMARTerTM PCR cDNA Synthesis Kit ovided by Clontech company.After recovery and purification in sections, the recovered 11-length cDNA were ligated to T-vector and transformed to DH5a. By cultured of onoclones,2410 positive clones were obtained. Selected 52 clones were sequenced. le average length of ESTs was 908bp. After BLAST comparison, these sequences were ghly homologous with the ESTs from Cicer arietinum, Medicago truncatula, Glycine ix and other plants. Construction of full-length cDNA Library of Caragana ermedia leaves under drought stress laid the foundation for the research of gene ;ource preservation and plant drought resistance mechanism.
