Scholars at home and abroad have payed attention to the halotolerant or halophilic microorganisms,just considering that they have the unnomal hereditary property metabolic mechanism, and physiological structure. They have been developed a resource with great value of utilization and academic research of microorganisms. This microorganisms’ unique physiological characteristics make them with excavating potential and research value of microorganism resources for getting the key new salt tolerance related genes, changeing of soil environment, traditional crops and so on.In this paper, we take a different approach to analysis one culturable moderately halophilic bacteria NEAU-ST10-9 from saline-alkaline soils in Songnen Plain and then consider the biodiversity and polyphasic taxonomy of it. Here we conside that 16 S r DNA, rpo B and gyr B sequences were used to analyze phylogenetic relationships and identif it in the family. As a result, we have gotten the bacteria NEAU-ST10-9’s information of phylogenetic relationships. It shows that the genus Planococcus may has a potential novel specie of NEAU-ST10-9.The taxonomic status of the the strain NEAU-ST10-9 by polyphasic taxonomy is described by us in this research, and the results just shows that strain NEAU-ST10-9 is a Gram-postive strain, and it’s aerobic, coccoid, but the colonies are circular, opaque, orange pigmented, convex smooth. Growth occurs at 4–50℃(optimum, 30℃), at p H 6.0–10.0(optimum, p H 7.0) and at 2–10 %(w/v) Na Cl(optimum, 4 % w/v). MK-7 and MK-8 are of the major menaquinones. Two kinds of saturated fatty acids C15:0 anteiso and C17:0 anteiso, and two kinds of unsaturated fatty acids C16:1ω7c alcohol, C17:1ω9c are the major fatty acids. The typical cell wall peptidoglycan of strain NEAU-ST10-9 is L-Lys-D-Glu.Two unknown lipids and phosphatidylethanolamine, diphosphatidylglycerol, phosphocholine,phosphatidylglycerol compose its’ polar lipids. The DNA G+C content is 45.2 mol %. Planococcus maritimus TF-9T is one of the most phylogenetically related species with NEAU-ST10-9, but it just shows gyr B, rpo B, 16 S r DNA gene sequence similarity values of 83.7%, 87.0%, 99.1% respectively with NEAU-ST10-9. The results of DNA-DNA hybridization assays showed 55±1、52±1、48±2、42±1、37±1、35±1、32±2%(all<70%)relatedness between Planococcus. maritimus DSM17275 T 、 P. rifietoensis DSM 15069 T 、 P. plakortidis DSM 23997 T 、 P. citreus DSM 20549 T 、P.maitriensis DSM 15305 T 、 P. Salinarum KCTC 13584 T 、 P. columbae DSM 17517 T with strain NEAU-ST10-9T respectively. Taking all of the factors above strain NEAU-ST10-9 is a novel specie of Planococcus. And the name is Planococcus dechangensis, NEAU-ST10-9T( =DSM25871T andCGMCC1.12151T).
