By the classic culture method and gradient dilution isolation method ,the strain JLT-01 which can grow optimally in culture media that contain 10-15% NaCl was isolated from the Jilantai salt lake located in Inner Mongolia. According to the classic phenotypic characteristics which include morphological observation,Growth Factor and the physiological and bio-chemical characteristics,homologous alignment and gradient dilution culture include 16S rDNA analysis, (G+C)content and the DNA-DNA hybridization, the moderate halophile strain was identified to belong to Marinobacter.It is Gram-negative, non-spore-forming, non-capsule-forming,non-flagellum-forming, rod shaped. Colonies are cream-coloured, smooth, circular to slightly regular. Optimal growth occurs in the presence of 10-12%(w/v) total salt,in the presence of 3% (w/v) Mg2+, at 37°C,at pH 7.5; The following compounds are utilized as sole carbon and energy sources: glucose, sucrose,galactose,The following compounds are not utilized as sole carbon and energy sources: fructose,maltose,,ribose,sorbitol,rhamnose,arabinose,mannitol,mannose,xylose,lactose.Methyl red and Voges–Proskauer are negative. Indole and H2S are positive.Simmons'citrate test is positive. Nitrate and nitrite are not reduced. Catalase and oxidase are produced,Tween 80 is hydrolysed, but casein,amylase,urease are not.Benzidine hydrochloride is positive,Gelatin liquefaction is negative.To determine the taxonomy position of the newly isolated moderate halophile bacterium JLT-01 in evolution history, 16S rRNA sequence homologous alignment and phylogeny analysis were performed between it and related species. The DNA-DNA hybridization results showed that the strain was not related at species level to the Marinobacter lipolyticus. Based on the data of polyphasic taxonomy,we propose to assign strain JLT-01 to a novle specie of the genus Marinobacter,for which we propose the nameMarinobacter Jilantaisum.sp.nov..The type strain is strain JLT-01.
