Cosmetic efficacy accessment includes in vivo efficacy accessment and in vitro efficacy accessment. In this study, salidroside was chosen as an active material to research the application of cell culture technique in cosmetic efficacy accessment.This study was divided into three parts:cell experiment, in vitro transdermal transport experiment, and in vivo experiment. In cell experiment, human skin fibroblasts were divided into control group, injury group, and salidroside group. To study the protection effect of salidroside, injury group was exposed to H2O2or UVB, salidroside group was pretreated with salidroside before H2O2or UVB treatment. Cell proliferation was detected by MTT assay; Gene expression and protein secretion of MMP-1was determined by RT-PCR and Western Blot; cell ROS and apoptosis was detected by DCFH-DA and Hoechst-PI.This study proves that salidroside has the capacity to protect human skin fibroblast from H2O2and UVB damage; salidroside can permeate through nude mice skin, and the cumulative permeating amount of salidroside has a positive linear correlation with time within0.5-8h; Gel containing salidroside obviously improve skin water content, increase skin elasticity and skin smoothness compared with blank group; Results of cell experiments were in agreement with that in vivo efficacy assessment proving that cell evaluation can be used as an important basis of cosmetic efficacy accessment.
