Isoetes occupies the aquatic to terrestrial series of habitat types,Scholars have done a lot of research in cytology, spore morphology,development, system classification and evolution, anatomy, genetics,molecular biology, ecology and biogeography, etc. Considering the fullinformation as to the origin of of each structure, embryonic developmentOntogeny process to some extent is simple and easy to repeat the evolutioncharacteristics of the ancestors of species and the evolution process,Combined with the different pattern of embryology arises from the differentgametophyte morphology, So reproductive biology research of Isoetes isdesperately needed. But this respect reported less in China. The main reasonis that material in the natural difficulties of endangered degree andtraining.Isortes orientalis grow in marsh area as Isoetaceae, Isoetes. Itdistribute in tining putuoshan peak nature reserve of fujian province andLishui city of SongYang in zhejiang province,after separating from I.sinensis Palmer,which has narrow growth range. I. orientalis is China’sunique wildlife species, confirmed by the international commission onbiological species named in2005as the world’s endangered plants,endangered levels for ER. Isoetes asiatica is Japanese endemic species,collected from lake piglet. I. orientalis and I. asiatica are produced byexcessive between species. Ater assessmenting of six Isoetes geneticdiversity and population in east Asia, they are listed as the new speciesin2006. Based on the above all, I ensure the research materials. I obsevethe selected embryos and gametophyte by semi-thin sectioning, research thesporophyte morphological by paraffin section method, compare the mega-spore、micropre in I. orientalis and I. asiatica by scanning electronmicroscopy (SEM) metgid, record the leaces diaphragm by confocal laser scanning microscope, abtain leaf blade information from the nail polishleather information mark method.By observing the spore germination and gametophyte mature to embryonicdevelopment process in I. orientalis and I. asiatica, I reported repro-ductive biology research in I. orientalis and I. asiatica for the firsttime,which for the purpose of exploring the reproductive growth rule,providing a reference for biodiversity machine, providing a detailed newmaterial for related froups of classification system and its related. Thispaper compares the walls of the layer, color, decorative pattern of I.orientalis and I. asiatica.The result shows that: I. orientalis and I.asiatica are in dark brown, the spore wall of microspore. But equatorialaxis of ridge in I. asiatica more prominent than that of I. orientalis’.I observed rotation of early embryonic development phenomenon in I.orientalis. In addition, This article also according to I. orientalisembryo in the process of root, stem, leaf and leaf tongue organs such asthe detailed record their occurrence, differentiation, embryo developmentwith other true fern, compares the similarities and differences of theresults further verified the root and leaf priority theory, and proposeswe think may be endangered reasons, based on the given Isoetes protectionrelated opinion.
