This topic is based on the experiments that different concertration of humanepidermal growth factor(hEGF) can promote Stylonychia division and morphologicalregeneration. Based on the theory of the CDK kinases compounds can adjust celldivision cycle and hEGF can interact with its receptor. Using monoclonal cultivationand artificial wound regenertion experiment,using silver protein staining,electronmicroscope and immunofluorescence experiments for two kinds of epidermal growthfactor in Stylonychia division and the role of wound regeneration process andimmunological mechanism is studied.The followings are the results of the study:1.Adopting the method of cell counting and combining with controlling cell cycle,comparing four different concentrations(0.1μg/ml,0.3μg/ml,0.5μg/ml,1μg/ml),hEGFand mEGF affect on culturing the monoclonal spine tail worm split the proliferation ofshort and long term. Founding that any concentration of hEGF can promote celldivision, and the mEGF can’t do in any concentration,even inhibiting populationgrowth. Then,further more comparing three different hEGF concentrations(0.1μg/ml,0.3μg/ml,0.5μg/ml) on promoting effect of cells division. After ten consec-utive cyclesconclused when the cocentration is0.3μg/ml,Stylonychia cell consecutives dividfastest.2.In order to further verify division the two EGF have different roles of Stylonych-ia and discuss the immunology mechanism of action,the author carried on the im-munofluorescence experiments. The results show that hEGF had strong immuneresp-onse,but mEGF and control groups showed no.There may be some mate rialsimilar to the human epidermal growth factor receptor in Stylonychia.Because of theEGFR can combine with hEGF,accelerating the speed of cells division.3.Combines immunofluorescence experiments and wound regeneration experim-ent, observed the different time after trauma, treatment group and control group inbody hEGF speed and immunofluorescence reaction intensity and the change of spatialdistribution, in order to investigate the wound healing process of hEGF on Stylonychiaand mechanism. Results show that the treatment group of insect body hEGF was faster than the control group, the fluorescence reaction can be observed immediately aftertrauma, and mainly concentrated in the wound site. Postoperative fluorescence reactionstrength reached maximum when2h, then gradually weakened, and spread to thecytoplasm and cell membrane area, to four hours to repair is complete; Control theinsect body1h after trauma is fluorescent expression, the strongest when4h, thengradually weakened. That Stylonychia of wound healing and indeed the number anddistribution of hEGF position has a close relationship.4.In order to further identify the hEGF mechanism of action on Stylonychia,wealso observed by transmission electron microscopy (sem) experiments the hEGF’s im-pact on Stylonychia ultranstructure.The results showed that:the control group and ex-perimental group in the membrane structure and submicroscopic structure of mitocho-ndria showed obvious difference,hEGF cultured body menbrane structure more clearl-y,mitochondria morphology fuller,more intensive mitochondria inside the crest.Proba-bly the hEGF promoted the cell bioological membrane updates,and indirectly contrib-uted to the mitochondria show young state.May also be hEGF directly affects the ene-rgy metabolism of cells,so as to promote the energy metabolism of core cell mitocho-ndria,development.The exact mechanism of biology remains to be further research.
