The normal progression of the cell cycle is the basis for the cell to maintain the physiological process.In the cell cycle,there are a variety of proteins that are periodically expressed and apoptotic as the cell cycle progresses,called cyclins.Different cyclins play different roles in the cell cycle.Studies have confirmed that some cyclin expression is abnormal in tumors,and clinical experiments have also proved that the expression of cyclin changes significantly before and after radiotherapy,indicating that cyclin can be used as a predictor and prognosis of tumor diseases.Therefore,it is very necessary to study the function and mechanism of cyclin.In this study,protozoa mode Stylonychia lemnae as materials,the gene sequence of protein cyclin9 Stylonychia genome database was cloned and analyzed to study the gene using PCR and RNAi technology in the floating The function of Stylonychia lemnae in the process of asexual reproduction.The results showed that: the gene belongs to the cyclin K type cyclin,which is located in the macronucleus,micronucleus and plasma membrane.The protein is regularly expressed during the mitosis and interphase of Stylonychia lemnae.The constructed RNAi vector can effectively silence Cyc9 gene expression.After the Cyc9 gene is knocked out,the Stylonychia lemnae in the interphase undergoes deformation,and the cell nucleus has obvious radial changes.The cell cycle is stagnated for more than 12 hours and does not continue to divide.After the RNAi is stopped,the cells can recover and continue to divide..However,the cells contracted into a round shape after Cyc9 knockout in the division stage of mitosis Cyc9,and there was no obvious change in the size of nuclei.This study is the first to explain the function and possible regulation mechanism of cyclin K protein in the process of asexual division of single-celled organisms,broaden the field of cyclin K function research,and has reference significance for the study of cell cycle regulation mechanism.
