Using high performance liquid chromatography coupled with electronspray ionization/triplequadrupole mass spectrometry (HPLC/QqQ-MS) to determine marine phytoplankton variation indifferent nutrient concentration seawater pond in a certain period.1. The method of pigment content analysis based on HPLC/QqQ-MS was established.Chlorophyll a, chlorophyll b, β, β-carotene, lutein, fucoxanthin and other pigment signatures wereselected for summarizing the mass spectrometry rule to identify pigments. And the mass spectrometrywas performed in SRM mode. The results showed that this method was sensitive, accuratereproducible. According the above method, ten algae(Heterosigma akashiwo(NMBRah03-2),Heterosigma akashiwo(NMBRah03-2-2), Karlodinium veneficum(NMBjah047-1), Prorocentrumminimum(NMBjah042), Nannochloropsis oceanic (NMBluh014), Chlorellapyrenoidosa(NMBluh015-1), Pleurochrysis sp.(NMBjih026-1), Prymnesium sp.(NMBjih029),Skeletonema costatum (NMBguh004-1), Thalassiosira weissflogii (NMBguh021) and Thalassiosirapseudonana(NMBguh005) have been investigated for comparing the pigments distribution.2. Using LC-MS together with algae formula to dectect algae in seawater ponds. Diatoms,Dinoflagellates, Chlorophyceae, Prasinophyceae, Chrysophyceae, Haptophyceae, CryptohayceaeCyanobacteria and Prochlorophytes were estimated in seawater ponds. Diatoms, Dinoflagellates andChlorophyceae were preponderant aglae. Chrysophyceae, Haptophyceae, Cryptohayceae andCyanobacteria were in low content. After adding nutritive salt, the community structure changed.thebiomass of adding N/P/Si (96/6/30μmol/L) pond>adding30μmol/L Si>non-nutritive salt addingpond. Diatoms and Chlorophyceae in adding N:P:Si pond increased, and the Chlorophyceae increasedobviously; The abundance of Dinoflagellate, Chrysophyceae and Haptophyceae decreased. The resultshowed that N/P/Si promote the growth of aglae, especially for Diatoms and Dinoflagellates. Theabundance of Diatoms in adding30μmol/L Si pond>adding N/P/Si (96/6/30μmol/L)>non-nutritivesalt adding pond. The result showed that Si promote the growth of Diatoms, and the growth inhibitionof Diatoms affected by interspecific competition in abunant N/P/Si pond.In a word, A rapid method detecting the structural determination of marine phytoplankton inseawater pond was established and provided a reliable tool for microalgae and the screening ofshellfish nutrition.
