It is now evident that most of the mammalian genome is transcribed and a majorproportion of the mammalian transcriptome comprises long (>200nt) noncodingRNAs (lncRNAs) that lack any protein-coding capacity and distinct from any knownfunctional-RNA classes. LncRNAs play an significant role in transcription andpost-transcription and regulating genomic imprinting, the X chromosome silence,epigenetic modification, cell cycle, alternative splicing, cell fate decision,reprogramming, cell apoptosis, oncogenesis and nervous system development.Although only a small number of long ncRNAs have been functionally characterized,many lncRNAs have been shown to be developmentally regulated and/or expressed intissues-specific and cell-specific manner, practically in central nervous system (CNS),suggesting that lncRNA play important roles in diverse neural developmentalprocesses.Long noncoding RNA (lncRNAs) are expressed widespread in mammaliancentral nervous system and have key regulatory and functional roles in nervousdevelopment. At present, lncRNAs in nervous system are investigated in whole brainor different neuroanatomical regions at multi-cell level, which would miss lowabundance lncRNAs expressed in a very limited subset of cell types because of theextraordinary cellular heterogeneity of the mammalian nervous system. Thus, it willbe important to undertake deeper and more focused transcriptomic profiling analyseson specific cells in order to reveal the full repertoire of lncRNAs in the adult mousebrain. In this work we fulfill the first genome-wide characterization of long ncRNAsat single cell level in the adult mouse subventricular zone (SVZ) of the lateralventricles. We identified and characterized50long ncRNAs, many of whichdisplayed cell specific expression. Our study provides the first systematicidentification of lncRNAs in the adult mouse subventricular zone at single cell level.
