Recently people put much more focus on label-free bio-detection. Electrochemical immunosensor is one of the many approaches. This sensing system is advantaged in fast-reaction, minimization, low cost and the small volume of the sample.In this thesis, a novel microcantilever-based electrochemical immunosensor is proposed, where a microcantilever is used as the working electrode. Co-detection of microcantilever deflection and electrochemical signals is explored. The mechanism of surface stress of the microcantilever under electrochemical potentials is investigated. Integration of such electrochemical system is explored.The achievement and innovation of this study are as follows:1. A novel detection method combining electrochemical detection with microcantilever is put forward. Corresponding experiment platform is constructed.2. A theoretical model of the immunosensor based on electrocapillary curve is build up. The simulated results agrees well with the reported experiment data.3. The effects of the electrochemical parameters in HClO4 electrolyte on the surface stress of the microcantilever is experimentally demonstrated. Research shows that the surface stress of the microcantilever is related to the surface charge density. The surface stress induced is reversible and repeatable.4. Such electrochemical system is used in PBS for study on the electrochemical detection and the surface stress of the microcantilever. The result indicates that the radius of the ion have an effect to the surface stress of the microcantilever.5. An on-chip electrochemical sensing system is innovatively designed. Fabrication of the chip of most steps is finished.
