Aptamers are artificial receptor, primarily DNA or RNA sequences that can bind with high affinity and specificity to low-molecular-weight organic or inorganic substrates or to macromolecules. Electrochemical immunosensor analysis is a method of the electrochemical and immunological techniques devices with rapid, sensitive, highly selective, easy to operate etc features.In order to study the aptamers and antibodies in the modified electrode specific detection of target molecules, we did some research about the GC electrode modified with gold nanoparticle in the following studies: (1) electrochemical aptamer sensor research was presented and the research on the electrochemical immunosensor a brief overview. Then the research proposal for this thesis was presented; (2) we developed a label-free electrochemical RNA aptamersor and doxorubicin can be used as an electrochemical indicator due to the strong affinity to the RNA double helix by intercalating between CG and GC of RNA, provides a new selection for theophylline detection; (3) we developed a mercapto labled electrochemical RNA aptamer,by measuring the electrical signal of ferrocene to achieve high sensitivity for detection of theophylline; (4) a novel bifunctional immunosensor based on a nano-Au film onto GC electrode has been constructed, used Hb to instead of BSA avoid the non-specific adsorption, make it not only realizes the determination of serum anti-HIgG levels, but also dramatically facilitate reduction of O2.
