[Objective]To prepare the serial tissue sections of lip and nose for establishing a visible virtual lip and nose model by the three-dimensional reconstruction software and study the morphological structure of philtrum and Labial arch. And the digital 3D model of lip and nose can supply basement for the study of morphology and the design, even the simulation of the operations of lip and nose.[Methods]2 adult lip specimens (A1; A2) and 2 fetal nose and lip specimens (B1; B2) were imbedded in the paraffin and serially sectioned with the thickness of 30μm. After the slices underwent the HE staining, the serial section preparatio images were pictured by digital camera and saved as the format of JPEG. Then the two-dimensional images were processed with Adobe Photoshop 6.0 and the three-dimensional model of nose and lip was reconstructed in computer with the 3D-DOCTOR Mimiscs and 3DMed software. Furthermore, philtrum of normal fetus and Labial arch of the normal fetus and those with cleft lip were sectioned, underwent HE and Masson staining, observed under microscope.[Results]139 and 258 pictures of adult serial slices were obtained, 266 and 252 pictures of fetal serial slices were obtained. The serial slices show the entire histological morphology of lip and nose as well as the philtrum. There are many decussate orbicularis oris muscle attached to the subcutaneous part of the philtrum, which play an important part in maintaining the structure of the philtral column. The thickness of the connective tissue under the Labial arch is smaller than those around. The anterior projection of the pars marginalis of the musculus orbicularis oris give rise to the normal Cupid's Bow.The visible nose and lip model was succeeded reconstruction with the medical 3D reconstruction software. The primary digital 3D model of lip and nose was established. The reconstructed lip and nose displays the histological morphology of the lip and nose and can be freely rotated, incised, restored. It supplies a good digital model for study and operation simulation such as the cheilorrhaphy.[Conclusions]We explore many experience of how to make the bigger tissue serial slices with paraffin method. The slices picture is clear and the color is vivid. The histological morphology of the philtrum guide the reconstruction of it during the cheilorrhaphy and the repair of the secondary deformity after the cheilorrhaphy.The method of 3D models reconstruction from serial histological sections of lip is available. Some experience of how to process big volume data and the reconstruct method was obtained. The digital 3D model possesses nice visible character which act as the explore of application of the model in medical field.
