The Ploy-enzyme Chain Reaction ( PCR in brief) is a classical technique for the molecules being augmented outside of body, in general, the augmentation process can be completed with performing a twenty to thirty circles reaction of high temperature disassembly, low temperature anneal, and middle temperature extension. In theory, one circulation can make the new gene copies rising about one time, so there would be 220 times gene copies of original after 20 circulations. In gene engineering, people can gain vast clonal growth copies of DNA through PCR technology, in order to perform qualitative analysis for gene.In this paper, a distributed temperature controlling system characterized by multi-position and pipelined treatment is put forward based on making an intensive study of theory of PCR, which includes twelve working positions can deal with twelve PCR processing of gene chip mostly at the same time. Controlling for nonlinear and uncertainty existing in the heated cavity of PCR system, a new FUZZY-PID controller is designed through simulation comparison among three kinds of PID methods. By setting the three parameters K_P, K_I, K_D on line according to the results of fuzzy reasoning and decision-making, the new FUZZY-PID controller has better response rate and stability property for temperature controling of heated cavity. Furthermore, the controlling theory based on single-chip microcomputer and implement methods of the hardware circuits designing are presented in thesis, including electric motor controling circuit, temperature sampling circuit and temperature adjusting driving circuit etc.; In order to supporting hardware, the writer selected VC++6.0 and ACCESS to develop manipulative program of upper, which is provide with serial communicating based on multi-thread, sampling and saving temperature, drawing temperature curve etc. The clock interrupt servicing program of lower is also presented, which can deal with FUZZY-PID algorithm.After a lot of performance tests, the results show that the multi-position pipelined PCR augmenting device meets the requirement of design in accuracy, stability and speed of temperature controling basically, the prototype according to research result is manufactured, which make necessary preparation for future development.
