This paper has designed and optimized an integrated digital microscope system, which can realize both auto-focusing and real time fusion. The system is mainly controlled by computer. It uses high resolution digital camera for microscope to obtain the image data and high subdivision step motor controller to rotate the step motor. Both the image data and the motor control commands are transferred through USB port.On the research of auto-focusing algorithm, the paper first analyzes existing auto-focusing algorithms. Based on this, a modified mountain climbing search method is embedded into DirectShow. Combining with our self-developing digital camera, the system directly does the search and control of the focal plane image on the bottom. The experiment results show that the modified algorithm has wider applicability and faster speed, and therefore has comparative practical value.Aimed at the shortcoming that microscope has less depth of field with higher magnification, the paper brings forward a software-based method to fuse multi-images around the focal plane. It can extract the clear image information in those partly-clear images, and then fuse them into one complete and clear image. By this, the depth of field of the microscope is extended. To realize seamless fusion, the paper has optimized the wavelet transform based fusion algorithm. This algorithm has greatly improved the fusion quality of images, and has definite theoretical significance and practical value.
