With the development of computer science and technology, Augmented Reality has gradually become the next generation technology of human-computer interface. By tracking camera's extrinsic parameters and intrinsic parameters in real time, Augmented Reality superimposes a computer-generated image onto the user's vision of the real world. It is able to provide a more natural user interface and establish a more realistic environment for applications. To date, many AR systems have been developed, for example, the system in which we can access to the observation of three-dimensional molecular structure and exploring the component of the gene chip. With computer graphics and interactive technology, multimedia technology and network technology, Augmented Reality has been widely used in the field of medicine, military, industries, entertainment and so on.This dissertation comprehensively introduces the conception, characteristics and an application of Augmented Reality. We analyze the architecture of general Augmented Reality system, its key technologies and the functions of several important modules, including image acquisition, image segmentation, 3-D registration, depth-testing, 3-D graphics rendering, and user interaction modules. We also analyze the related algorithms, e.g. RAC camera calibration algorithm based on Tsai's camera model. Then we explain in details the implementation of key technologies in the camera calibration module, the 3-D models rendering module, the animation-rendering module, the collision detection module, and the multi-media technology module of our prototype system. At last, we present a concept of marker's directional property; furthermore, we realize a practical solution for extending the marker identification's space based on this concept in order to improve the stability of the common Augmented Reality system. Based on prototype system, we developed a virtual genetic laboratory ,which is a part of Digital Science and Technology Museum , and Cooking Huaiyang Cuisine system of Digital Culture Museum.
