Intracellular free calcium ([Ca2+]i) is one of the most important second messengers in living cells. [Ca2+]i regulates many physiological activities. It is important to research the activity of Ca2+ inside and outside of the cells. Fluorescence microscopy has many advantages, such as high temporal resolution and low toxicity to the cells. So the fluorescence microscopy is an important and significative method to study in living cells. The primary work mentioned in this thesis is to develop the control system in the fluorescence microscopy system to detect [Ca2+]i in living cells. The main function of the control system is to control the Mirror Positioning System of monochromator to emit monochromatic light, which is set by user. To ensure the safety in xenon lamp room, the temperature monitor circuit base on ratio method was designed in this system. And the principle of detect the temperature by Pt100 sensor is given. Organic Light Emitting Display, OLED is used for displaying the state of the monochromator.Intelligent function is very important in modern electronic equipments. For improve the intelligent level of the control system, the manual controller was designed. The microprocessor in the manual controller is SST89E564, which have in application programming function. The manual controller provides an interface of man-machine conversation for experimenter. Using this interface, user can set parameters of emitting monochromatic light. The manual controller communicates with microprocessor FX2 in the control system by the RS232 protocol, and then FX2 translated digital data into analog signal that control the Mirror Positioning System. At last, how to use the monitor-51 protocol of Keil uVision2 to develop the program of microprocessor was discussed. The result of test was given in the end.
