Objective To develop a model of acute intracerebral hemorrhage(ICH) and investigate the effects of Neuregulin-1β(NRG-1β) on the expressions of complement C3 and C4, evaluate treatment with NRG-1β on edema extent. To investigate the neuroprotective effect of NRG-1β on the inflammatory response following ICH.Method ICH was induced by stereotaxic intrastriatal injection of collagenase VII(0.5U) in adult male SD rats, 20 randomly selected rats received an equal volume of saline infusion as a shamoperation group. 88 successful ICH model rats were randomly divided into a control group and a treatment group(n=44) according to drug administration. On the third day, four rats were taken from each group, cerebral edema was assessed using dry and wet right hemisphere weighing. Then the rest were evenly subdivided into 1 d, 2 d, 3 d, and 5 d subsections. Rats were administered Neuregulin-1β(4 μg/kg) or equal amounts of vehicle intraperitoneally two hours after ICH. The expressions of C3 and C4 proteins were determined by immunohistochemical staining and Western Blot analysis, neurological dysfunction was evaluated by means of the 28-point neurologic function score.Results Compared to vehicle, the expression of C3 and C4 was escalating with the prolonging of the hemorrhage time and reached the peak at day 3 with a subsequent decline at day 5 in both treatment and control groups. Although both groups showed a similar fluctuation curve, compared with the control group, treatment with NRG-1? resulted in a dramatic decrease in the level of expression of C3 and C4(P<0.05), relieved cerebral edema and reduced neurological dysfunction.Conclusion NRG-1β can reduce the expression of C3 and C4, relieve cerebral edema, reduce neurological dysfunctions in rats after ICH, thus inhibits the development of inflammation by influence complement cascade, improves the microenvironment of perihematoma. Therefore, suggesting a potential protective role for NRG-1β in ICH.
