| Objective1ã€To make sure (explore) the mechanism of HSG (hyerplasia suppressor gene, HSG) intervening airway remodeling of COPD (Chronic Obstrutive ulmonary Disease, COPD).2ã€To explore the mechanism of the Miao medicine of cough and phlegm (Myrsinaceae, Lespedeza cuneata equivalent) inhibiting airway emodeling of COPD and lay the experimental theoretical foundation for clinical research.MethodsThe healthy male SD rats were divided randomly into normal group, blank group of rats with COPD, Miao medicine of cough and phlegm (Myrsinaceae, Lespedeza cuneata equivalent) gavage treatment group and intramuscular treatment group. we established COPD model by using the method of smoking cigarettes and injecting Lipopolysaccharide into trachea of SD rats, and treated those rats with Miao medicine and dexamethasone, respectively. We detected the levels of PDGF (Platelet derived growth factor, PDGF) and TGF-β1(Platelet derived growth factor, TGF-β1) in BALF (bronchoalveolar lavage fluid, BALF) by EL1SA (enzyme linked immunosorbent assay, ELISA) Then, fibroblasts were extracted from small airways of rats.which were used to primary culture.we used western blotting to detect the protein expression of rHSG (rats hyerplasia suppressor gene, rHSG) and used RT-PCR (reverse transcription-polymerase chain reaction, RT-PCR) to detect the mRNA expression of rHSG in fibrocytes of airway to study the expression of rHSG in COPD fibroblasts which were treated by Miao medicine of cough and phlegm and the correlation between that and PDGF, TGF-β1.Results1ã€rHSG and mRNA in normal group was over expression and showed low expression in COPD model group, while the expression of PDGF, TGF-β1were contrast with the former in both groups. the difference between two groups had significant statistical significance(P<0.01).2ã€The expression levels of rHSG and mRNA in COPD model treatment group (gavage treatment group and intramuscular treatment group) were higher than COPD rat model,which had significant statistical significance(P<0.01). However, PDGFã€and TGF-β1COPD model treatment group was low expression than that in COPD model group.the difference between two groups had significant statistical significance (P<0.01).3ã€The expression of rHSG was negatively correlated with that of PDGF, TGF-β1which had significant statistical significance(P<0.01).Conclusions1ã€HSG inhibits airway remodeling of COPD by down-regulating PDGF and TGF-β1.2ã€Up-regulating HSG and inhibiting the expression of PDGF and TGF-P1is one of mechanisms Miao medicine of cough and phlegm (Myrsinaceae, Lespedeza cuneata equivalent) intervene airway remodeling in COPD... |
