Tuberculosis is one of the main enemies of human health. Chemical treatment is the most common treatment method of Tuberculosis, but this method occurs frequently in some side effects and the drug resistance problem is becoming more and more serious. Therefore, discovering new anti tuberculosis drug were became one of the more important task. In recent years, the cases of double infection of HIV virus and Mycobacterium tuberculosis have rised rapidly, human beings are facing the dual threat to HIV epidemic and drug-resistant tuberculosis. So, research the anti tuberculosis drug brook no delay. The researches of natural anti-tuberculosis drugs have received great attention from domestic and foreign scholars at present.The earliest anti streptomycin TB drugs were extracted from actinomycetes, after decades of screening mining it seems difficult to antibiotic producing bacteria in the soil from the new found common. In recent years. So actinomycetes are still an important source of research and development of anti tuberculosis drugs.In this paper, we combined with the unique geographical landforms Guiyang City, more than 40 strains isolated from Huaxi District, Guiyang city surface freshwater bottom soil of the Communist Party of china, after the surface morphology of macroscopical character, combined with the same strain, a total of about 20 strains of actinomycetes, and established the Huaxi area surface freshwater bottom soil actinomycetes sample library. For anti tuberculosis activity test of actinomycetes in sample library, and G2 was obtained with activity against Mycobacterium tuberculosis. Further extracted with petroleum ether and ethyl acetate, we found that the extracts of ethyl acetate have the activity against Mycobacterium tuberculosis, and the MIC value is 400μg/ml. To detect G2 strains, combined with 16 S rRNA sequence alignment results, strain G2 was identified to Streptomyces albogriseus. And we firstly discover that Streptomyces albogriseus has the activity against Mycobacterium tuberculosis.Secondly, we use the medicinal plant resources of Guizhou unique and diverse and establish a plant sample library. 32 plants were detected activity against Mycobacterium tuberculosis and found that Achyranthes bidentata and Dushan fissistigma oldhamii have the activity against Mycobacterium tuberculosis. According to reports, Achyranthes bidentata entire grass can be medicinal, and have antiphlogistic hemostatic effect, but there are few reports of Achyranthes bidentata in the application of anti Mycobacterium tuberculosis. Fissistigma oldhamii is mainly used to treat rheumatic arthralgia, back pain, abdominal pain and abdominal distension and traumatic injury, and there are already a part of companies of fissistigma oldhamii production of Zuanshanfeng syrup.We carried out the activity of anti Mycobacterium tuberculosis tracking separation of Achyranthes bidentata and Dushan fissistigma oldhamii, separated NX-01 and GF-02 from Achyranthes bidentata and Dushan fissistigma oldhamii, they all have the activity against Mycobacterium tuberculosis. And we measured NX-01 MIC value to 400μg/ml, GF-02 MIC value to 100μg/ml. Compound NX-01 and GF-02 with INH respectively and isoniazid for joint action and the combined effect is synergistic effect. The MIC of NX-01 become 200μg/ml and the MICof GF-02 become 50μg/ml. This method can reduce the degree of isoniazid resistance, so as to reduce the clinical dosage, reduce the adverse effect incidence.Guizhou has a large number of Achyranthes bidentata and Dushan fissistigma resources, this project has studied the pharmacological action of anti bacillus tuberculosis of Achyranthes bidentata, Dushan of fissistigma oldhamii and Streptomyces albogriseolus, for the will of Achyranthes bidentata, Dushan of fissistigma oldhamii developed into drugs and laid a solid foundation. This item separation anti Mycobacterium tuberculosis active monomer purified from Achyranthes bidentata and Dushan fissistigma oldhamii, will further structure modification, on the findings have important implications for new anti tuberculosis drugs.
