Objcetive:The study use the methods of the proteomics to dicuss the influence of the differences expressed of the left ventricular muscle protein grou in the small intensity exercise. Screening the target protein of the heart in the sports response and adaptive.Finding the new protein which related small and medium-sized intensity aerobic exercise from proteomics of quantitative and qualitative. Providing the evidence and theoretical guidance for the exercise of cardiovascular disease and chronic rehabilitation treatment.Methods:20 rats were divided into rest and exercise group in random. The exercise group was set up to do treadmill training with medium and small intensity exercise and then killed to extract the total protein of left-ventricular muscle for two dimensional gel electrophoresis separation. The spots in differential expression volume more than 5 times are selected as alternative spots for mass spectra (MS) identification. The gels maps were analyzed with the Bio PD quest image analysis software to choose variable proteins and the chosen proteins were identified by ULGRAFL-FLEX-TOF/TOF, detect the gene mRNA transcription of target protein With RT-PCR.Results:①After 8 weeks' training, the index of heart weight of exercise group increased 32% compared with that of the control group, the rat heart of the test group present morphological change.②In the exercise group left ventricular muscle protein expression differences is raised≥5 times 12 protein sites, The mass spectrum identification corroborated 7 protein and a unknown protein. Respectively glutathione peroxidase 1, after motion expression is raised 5.2 times; cAMP dependence protein kinase type alpha, after adjusting the movement lowed 5.8 times.The expression of Cytochrome b-c1 complex subunit Rieske, mitochondrial raised 14.2 times after sports;β-enolase's expression raised 20.5 times after sports;The expression of Voltage-dependent anion-selective channel protein 1 raised 5.9 timesafter sports; Inner membrane protein, mitochondrial's expression is lowered the 7.2 after sports;The rhophilin, Rho GTPase binding protein 2's expression is lowered 9.6 times,and a unknown protein,molecular weight is 138.478 KDa, which expression is lowered 12.1 times.③The results of RT-PCR shows that theβ-enolase's mRNA relative density of light is 2.25±1.41253 in control group,in the sports is 1.76±0.98471.β-enolase's mRNA expression of quantity has cut trend, and mass spectrum identification results are not consistent. cAMP dependence protein kinase type alpha adjustment and base of the relative density of light mRNA 0.18071±0.68 in the exercise group,0.32452±for 0.93, compared with the control group, the sports cAMP dependence protein kinase type alpha adjustment they express quantity, mRNA and mass spectrometry appraisal result in cAMP dependence protein kinase type alpha adjustment and expression in exercise is not consistent cut.Conclusions:①After long aerobic exercise, left ventricular muscle proteome has changed significantly.②Rats right ventricular muscle protein occurred significant differences after eight weeks movement③Cardiac energy metabolism level and Contractility was enhancement, rat cardiac appear good adaptability changes during the moderate and small intensity aerobic exercise conditions.④Protein level and mRNA level must not positively correlated, The influence of movement can be adjusted from various mechanisms.
