Objective:This research discusses the influence of the4weeks aerobic exercise to the differential expression of the rat heart muscle protein group,to screen target protein point in the atrial muscle which is closely related to Aerobic exercise stress and study the mRNA expression level of part of the target protein point. To provide theory basis and new ideas for further revealing the occurrence and development of the movement of heart and the exercise rehabilitation research of the nationwide fitness campaign and Chronic heart disease.To provide theory basis and experiment basis to the continue study of the movement of heart of proteomics.Methods:20SD male rats are divided into sports group and control group randomly according to their weight weight and exercise ability(n=10). Using the way of treadmill exercise and after adaptive training, copying24m·min-1x40min athletic intensity load a continuous training4weeks aerobic exercise experimental animal model (load intensity is equivalent to60to70%VO2max). Six hours after the last time of the training, the rats of the experimental and control group of movement executed with Anesthesia, and extraction of the muscle tissue protein the heart, the two-dimensional gel electrophoresis technology (2-DE) on the protein separation, on2-DE atlas to carry on the analysis, and application series tofms (ULGRAFL-FLEX-TOF/TOF) on one of them to express quantity change raised more than5times and down to below1/513goals protein points mass spectrum identification, with drug-the polymerase chain reaction (RT-PCR detection technology to seven goals of protein gene expression level for transcription of the mRNA detection.Results:(1) after four weeks’aerobic exercise, compared to the quiet control group, the heart weight of rats increased1.7%, heart heavy index increased32%(P<0.05);(2) in2-DE electrophoresis, the campaign group detected370+35protein points, quiet control group detected422+7protein points.After exercise,there are53differential expression protein points,included29whose expression raise more than two times,23whose expression down to more than two times and5whose expression raise to more than five times and8whose expression down to more than five times. After exercise,the expression of Atrial muscle protein group of rats occured differential changes obviously.The mass spectrum analysis of the13protein points whose expression raise to more than five times or down to more than20%identified successfully11kinds of protein and an unknown protein whose molecular weight was54KDa,.These proteins were similar transcription catalytic protein alpha, pyruvate dehydrogenase E1α1, mitochondrial monkshood acid hydration enzyme, methyl malonic half aldehyde dehydrogenase [acyl], whereas hyperthermia accelerated the base, mitochondrial protein flavin two hydrogen lipoic acid dehydrogenase, disulfide protein isomerase an A3, glutathione synthase, isoamyl usions coenzyme A dehydrogenase, cytoplasm malata dehydrogenase, silk protein kinase activity crack element3.(3) RT-PCR detection results:the inRNA relatively bare density values of methyl malonic half aldehyde dehydrogenase and cytoplasm malata dehydrogenase of rats artrial musles in the exercise group was lower than those in the control group (P<0.05); the mRNA relatively bare density values of Mitochondria two hydrogen lipoic acid dehydrogenase, glutathione synthase mitochondrial monkshood acid hydration enzyme and protein disulfide key was lower than those of the control group (P>0.05); the mRNA relatively bare density values of Isoamyl ene coenzyme A dehydrogenase was higher than that in control group (P>0.05). The expression level of mRNA of the mitochondrial two hydrogen lipoic acid dehydrogenase, glutathione synthase and cytoplasm malata dehydrogenase was corresponding with mass spectrometry appraisal of the protein expression level consistent, all downregulation.Conclusion:(1) after four weeks aerobic exercise, atrial muscle protein group of rats has a significant differential expression.(2) the selected target protein points,such as pyravate dehydrogenase E1α1, mitochondrial monkshood acid hydration enzyme and silk protein kinase are related to raise the level of Myocardial cell energy metabolism and Sports prompted myocardial cell hypertrophy proliferation (3) after4weeks aerobic exercise,the gene expression level and protein expression level of target protein is not identical.
