Purpose:Exercise has been shown to induce oxidation stress. Measurement of 8-iso-PCF2αis the most reliable approach to assess oxidative stress status in vivo, providing an important tool to explore the role of oxidative stress in the pathogenesis of human disease.Acting as an antioxidant, lycopene has been attached importance in recent years. The purpose of this present study is to discuss the usefulness of quantifying 8-iso-PGF2αas an index of oxidative injury in association with exercise and the 8-iso-PGF2αin urine as a non-invasive approach in exercise area.Through analyzing the influence of the lycopene on 8-iso-PGF2αand antioxidant defenses to rest and exercise,we can supply a certain academic foundation for lycopene reasonable use. Methods:The male athletes of 16 judoka cycled for 45min at approximately 60%V02max and followed by 90%V02max to exhaustion immediately after two-week of placebo(C group) or lycopene(L group) supplementation.Blood was taken in the state of quiet before and after medication, submaximal and high-intensity exhaustive exercise, and after 1h of recovery.Urine was taken at rest,high-intensity exhaustive exercise, after 1h of recovery and 2h of recovery.Serum oxidant stress, antioxidant defenses and urine 8-iso-PGF2αwere examined.Results:1.Serum oxidant stress, antioxidant defenses and urine 8-iso-PGF2αwere unaffected by two-week of lycopene supplementation. 2.Serum 8-iso-PGF2αconcentration in the C group was significantly higher than that in the L group after submaximal exercise. It decreased after one hour recovery.Serum SOD concentration in the C group was significantly lower than that in the L group after exhaustive exercise. Urine 8-iso-PGF2αconcentration in the C group was significantly higher than that in the L group after submaximal exercise.3.Serum GSH-Px,T-AOC concentration in the C group significantly increased after exhaustive exercise and decreased after one-hour recovery.GSH-Px concentration in the L group was significantly higher than that in the C group.Conclusion:1.The serum 8-iso-PGF2αconcentration significantly increased after exercise. It peaked immediately when athletes stop excising, and decreased gradually. It provides an useful tool to define the oxidant stress of exercise.2.8-iso-PGF2αin urine was significantly increased after two hour recovery.Measurement of 8-iso-PGF2αin urine provides a reliable non-invasive approach to assess lipid peroxidation invivo after exercise.3.These results suggest that two-week of lycopene supplementation may be not effective at rest, but it blunts the lipid peroxidation response to acute bout of exhaustive exercise independently of changes in oxidative stress.
