Hemojuvelin was found in 2004, is an important regulatory protein of iron metabolism. The important role of HJV was to regulate the expression of Hepcidin,and Hepcidin plays an important role in regulating iron absorption in the intestine.s-HJV inhibited the expression of Hepcidin,m-HJV promoted the expression of Hepcidin, the two HJV Interacted to maintain the dynamic balance of Hepcidin in the liver.More and more scholars are concerned about changes in iron metabolism. Especially the mechanism of exercise-induced hypochrosis was extensive and in-depth study. However,on the HJV dynamic expression in the formation of the exercise-induced hypochrosis needs further study.Therefore,we study the expression of HJV during the exercise-induced hypochrosis in rats,in order to reveal the mechanism of exercise-induced hypochrosis, then easily to reflect the iron metabolic status of body timely in the training practice and prevent the occurrence of exercise-induced hypochrosis.Methods: 36 male Wistar rats were randomly divided into 2 groups:control group (CG) and exercise group (EG).CG group was divided into three sub-groups by post-exercise 3 weeks,4 weeks,5 weeks;EG group was divided into three sub-groups by post-exercise 3 weeks,4 weeks,5 weeks.The rats of treadmill running with different intensities and progressive load in EG.Exercise training consisted of running on a motorized treadmill at 30 m/min,the treadmill was always kept at 0% inclination.The rats were trained once a day during the first 2 weeks and then twice a day for the next 3 weeks.The training time was increased by 2 min each session for 6 days/week.The last run time was 95 min.The expression of blood in the formation of exercise-induced hypochrosis were determined by an automatic blood analyzer, Serum samples were analyzed for circulating levels of HJV by means of enzyme-linked immunosorbent assay(ELISA).The expression of gastrocnemius s-HJV mRNA,hepatic m-HJV and hepatic Hepcidin mRNA were examined by reverse transcriptase-polymerase chain reaction (RT-PCR).Results:(1)After 4-week running,The levels of body weight,hemoglobin(Hb),and MCHC were lower in EG than in CG(P<0.05),RDW was higher in EG than in CG(P<0.05). After5-week running,The levels of body weight,Hb,MCHC were significantly lower in EG than in CG(P<0.01),RDW was significantly higher in EG than in CG(P<0.01).(2) After5-week running,the amount of s-HJV and the mRNA expressions of s-HJV in gastrocnemius were lower in EG than than in CG(P<0.05). The amount of s-HJV mRNA in gastrocnemius were lower in 5EG than than in 3EG (P<0.05).After 3-week running,the mRNA expressions of hepatic m-HJV mRNA was lower in EG than in CG (P<0.05).There was no significant changes of the mRNA expressions of hepatic m-HJV mRNA between 4-week EG and CG.After 5-week running,the mRNA expressions of hepatic m-HJV was significantly higher in EG than in CG (P<0.01). The mRNA expressions of hepatic m-HJV mRNA was significantly higher in 5EG than in 3EG (P<0.01).The mRNA expressions of hepatic m-HJV was higher in 5EG than in 4EG (P<0.05).(3)After 3-week running,the mRNA expressions of hepatic Hepcidin was lower in EG than in CG (P<0.05).There was no significant changes of the mRNA expressions of hepatic Hepcidin between 4-week EG and CG.After 5-week running,the mRNA expressions of hepatic Hepcidin was significantly higher in EG than in CG (P<0.01). The mRNA expressions of hepatic Hepcidin was significantly higher in 5EG than in 3EG (P<0.01).The mRNA expressions of hepatic Hepcidin was significantly higher in 5EG than in 4EG (P<0.01).Conclusion: The strenuous exercise could decrease serum s-HJV and gastrocnemius muscle s-HJV mRNA then the expression of hepatic Hepcidin mRNA was increased.The high expression of Hepcidin mRNA may be one of the most important reason for the formation of the exercise-induced hypochrosis.
