Objective:Through establishment the model of rat of diabetic retinopathy, observed the effect of angiopoietin-1 (Ang-1) in vascular endothelial growth factor (VEGF) and diabetic rat retinal neovascularization by the vitreous cavity injection, and provided experimental evidences for the treatment of diabetic retinopathy using Ang-1.Methods: 48 healthy SD rats, 16 of them were randomly selected as normal control group(A group) and were not treated. Another 32rats were injected of 10% solution streptozotocin (STZ) through lower left abdominal with the dose of 65mg/kg. After injection, the blood glucose was detected in 2d, 7d, 10d. Rats with the postprandial blood glucose levels greater than 16.7mmol / L and maintained a stable blood sugar are the diabetes model. Feeding after 4 months checked the establishment of diabetic retinopathy in rats by fundus fluorescein angiography. Then, the model rats were randomly divided into two groups (n = 16), positive control group(B group)and the Ang-1 treatment group(C group). Positive control group (B group)were injected phosphate buffered saline (PBS) 5μl through the vitreous cavity, Ang-1 treatment group(C group)were injected Ang-1 5μl of a concentration of 160μg/mL. Three days later, repeat the above treatment once again. Continue to monitor three days,rats in each group were sacrificed and the eyes were resected, and detected the expression of VEGF in each group retina by immunohistochemical, comparing the number of endothelial cells by conventional HE staining, which were breakthrough the internal limiting membrane.Results: (1) Compared with the normal control group(A group),the VEGF expression of positive control group (B group)and the Ang-1 treatment group(C group)was significantly increased, (P<0.05), the comparison of optical density values has statistical significance (P<0.05); the VEGF expression of Ang-1 treatment group(C group)was significantly reduced compared with the positive control group(B group), the comparison of optical density values has statistical significance P<0.05). (2) Compared with the normal control group(A group),the number of breakthrough internal limiting membrane cells of positive control group(B group) and the Ang-1 treatment group(C group)was significantly increased, and have statistical significance (P<0.05).The number of breakthrough internal limiting membrane cells of the Ang-1 treatment group(C group)was significantly reduced compared with the positive control group(B group), and have statistical significance (P<0.05).Conclusion: When Diabetic Retinopathy occurs,the rat retina will produce a large number of VEGF. Injection the Ang-1 into the intravitreal can inhibit the expression of VEGF of STZ-induced diabetic retinopathy in rats, and reduce the generation of retinal neovascularization.
