Objective: To explore the expression and relationship of 14-3-3σ,ER(estrogen receptor),PR(progesterone receptor) in human uterine leiomyoma tissue.Methods: The specimens were choosed from 80 cases of uterine leiomyoma and homologous normal myometrium obtained at hysterectomy from eighty reproductive women who had not received steroid hormone treatment.The expression of 14-3-3σwas measured by reverse transcription - polymerase chain reaction (RT - PCR) and western-blot in 20 cases of those specimens.By means of immunohistochemistry,the 14-3-3σ,ER,PR expression level were detected in 80 cases of those specimens,circulating levels(14-3-3σ,ER,PR)were also analysised.The experimental datas were obtained by ImageTool software. Statistical analysis was performed using SPSS for windows 15.0 software.Results: In uterine leiomyoma tissue,14-3-3σ,ER,PR positive expression rates were 40%,85%,88.75%,respectively.Compared with homologous normal myometrium,the expression of 14-3-3σwas down–regulated in uterine leiomyoma tissues(P<0.05).The expression of ER,PR in uterine leiomyoma tissues were obviously higher than those of homologous normal myometrium (P<0.05).The expression of 14-3-3σ?was negatively correlated with ER and PR(ER:r=-0.235 ,P<0.05;PR:r=-0.275 ,P<0.05).Conclusion: The lower levels of 14-3-3σclosely related to the pathogenesis of human uterine leiomyoma by impacting the expression of ER and PR.It is significant that the combination of detection of 14-3-3σand ER,PR in the pathogenesis of human uterine leiomyoma.
