Objective : Observe the effect of NaoTaiTong-granule (NTT-granule) treatmenting rat model with cerebral ischemic stroke, and initially explore treatment mechanism of NTT-granule on cerebral ischemic stroke.Methods:To select clean grade 120 SD rats with similar weight,the half rats were male,the rest were female.They were divided four groups: sham-operated group, placebo group, experimental group, positive control group.â‘¡Four groups of rats were fed with high fat diet for 10 days, except for sham-operated group, the rest three groups were made middle cerebral artery occlusion (MCAO) model by using thread method basing on relevant documents.â‘¢After the success of modeling,all the rats were treated with corresponding drugs and NS for 28 days.â‘£After treatment,at 7d, 10d, 14d, 21d, 28d time points,we observed behavior change of rats and neuronal changes of rat brain tissue by HE staining.We detected expresstion of Slit2 and VEGF in rat brain tissue by immunohistochemistry.Result:â‘ At different time points after treatment, through neurological function was valued by Longa tests, we found that the score of experimental group rats were lower than positive control group,and the two groups existed statistically significant difference(P<0.05).â‘¡We could find positive cell expression of Slit2 protein in rat brain tissue at different time points after ischemia, and positive cell expression of Slit2 protein peaked at 7d,and it was more than 10d, 14d, 21d, 28d.Positive cell expression of slit2 protein of experimental group was more than any other groups, including placebo group and positive control group,they existed statistically significant difference(P<0.05).â‘¢At 7d, 10d, 14d time points, the VEGF protein content of experimental group were more than positive control group, they existed statistically significant difference(P<0.05). At 21d, 28d time points, the VEGF protein content of experimental group were more than positive control group ,however, they did not exist statistically significant difference(P>0.05).â‘£Pathological slice showed: necrosis area of the experimental group and the positive control group were all less than the placebo group at 7 day, we found that the capillaries around ischemic area all increased at 21 day, experimental group and the positive control group did not exist significant difference.Conclusion:After focal cerebral ischemia in rats, the expression of Slit2 and VEGF increased,but they increased significantly because of taking drugs, the experimental group was much more obvious than the positive control group.It told us that NTT-granule could promote the rehabilitation of limb function on rats ,improve the symptoms of neurological deficits and reduce infarct,by promoting axon growth and branch formation of hypoxic-ischemic in rats brain tissue and the promotion of vascular endothelial regeneration and the formation of new blood vessels.
