Rhizoma Paridis,or Paris polyphylla Smith var.chinensis Hera, is the general term for Liliaceae Parisand and is first recorded in Shennong's Herbal. The traditional Chinese drug Paris is the dried rhizome of Paris, most of which grows in southwest provinces in China. It has been a long history since Paris was used as medicine.There are lots of effecs,for instance,heat-clearing and detoxicating,detumescence and odynolysis, liver cooling and convulsion arresting. It is used to cure ulcerative carbuncle, venomous snake bite, pain and hyperspasmia and so on.Recently, with deeper study,the value of Rhizoma Paridis as medicine has been highly thought. But at present there are few reports about the study of Paris, mary studys focused on resource,pharmacognosy,chemistry and pharmacology.Purpose:To observe the effects of the active ingredients of the plant Paridis saponin I;and investigate on the invitro anti-proliferation of Bel-7402 cells, and the possible mechanisms.Method:By addition of different concentrations of Paridis saponinⅠdealing with the cell Bel-7402 at different times, MTT assay was used to exam the status of cell proliferation.Changes in cell morphology were observed under optical microscope.By culturing of Bel-7402 cells with Paridis saponinⅠat the concentration of 1.25μg/ml at different times,we detected apoptosis after propidium iodide (PI) staining by flow cytometry (FCM). Agarose gel electrophoresis was used to observe the apoptotic changes.Immunohistochemis--try was used to detect the expressions of bax, bcl-2 and caspase-3 proteins after the addition of Paridis saponin I.Results:It was observed that Paridis saponin I could inhibit the proliferation of the cell Bel-7402 within the dosage from 0.3125 to 5μg/ml and the effects were in a significant dose-dependent manner; It could induce apoptosis of the Bel-7402 cells. By FCM, apparent peak of apoptosis could be observed at the concentration of 1.25μg/ml at 12h,24h and 48h:the longer the time, the higher the rate of apoptosis. It was observed that DNA fragments significantly increased;The expression of apoptosis-related proteins bax and caspase-3 increased, while bcl-2 protein decreased with the precenceof Paridis saponin I at the concention of 1.25μg/ml at 24h. Conclusion:Paridis saponinⅠcan inhibit the proliferation of Bel-7402 cells and induce apoptosis.It can increase the expression of Bax and caspase-3 proteins, and reduce the expression of bcl-2 protein.Paridis saponinⅠmay play an anti-tumor role by affecting Bax and bcl-2 protein expression to activate caspase-3 pathway.
