Objective To examine the effect of arsenic trioxide ( As2O3) to ovarian cancer cell line SKOV3. Methods Methyl thiazolyl tetrazolium (MTT) assay was used to establish the dose-effect curves. The morphological changes of the cell were observed by transmission electron microscope. The change of cell cycle was analyzed by flow cytometry. The activity of caspase-3 was studied by flurometric immunosorbent enzyme assay (FIENA) . Result The inhibition of As2O3 to the growth of SKOV3 was dose-dependent. After 24 hours of exposure to As2O3 , SKOV3 cell line presented some morphologic features of apoptosis , including cell shrinkage , chromatin condensation and formation of apoptotic bodies. Some typical subdiploid peaks before Go/Gi phase were observed while the cells were arrested in G2/M phase. As2O3 activated caspase-3 and caused apoptosis. Conclusions As2O3 can inhibit growth of SKOV3 and induce apoptosis of it.
