Objective:Exogenous fever in children is a period of common diseases and frequently-occurring disease in children, especially in early childhood is most prevalent in the pediatric acute infectious diseases top the list. Exogenous fever in children with rapid onset, short course, communication rapid. It should be actively treated. Its main pathogens are certain viruses and bacteria have disease. In the current clinical use of antipyretic drugs in Western medicine certainly effective but oral medication there are mostly gastrointestinal reactions at the same time most drugs have strong promoting sudation has certain influence on pediatric rehabilitation. The subject of yeast-induced fever in rats produced animal models of rat fever in order to aspirin as a positive control drug by observing ShuJieTuiRe decoction on rat model of fever in general using immunohistochemical detection of rat medial hypothalamus preoptic area and median preoptic area PGE2,cAMP levels further explore the treatment of children with Exogenous this side effect and mechanism of antipyretic fever to prove the reasonableness of its group of parties in order to further guide clinical application.Method: The experiment included two parts. Healthy male Wistar rats that weighed about 180g to 200g were adopted in our research.(1) Effect on body temperature of febrile rats induced by yeast: Before the experiment, we made the rats adapt surroundings for three days. They could move, eat and drink freely, and they measured body temperature twice every day. During the experiment measured body temperature three times in the same way. The interval was half an hour. The rats whose fluctuations in temperature exceeded 0.3℃were rejected. Counting the average value of the temperature regarded as the basic body temperature. The rats were divided into five groups. Except for Normal group, the rest rats were injected 20% suspension yeast in the subcutis of back by 10ml/kg. After eight hours the rats whose body temperature heightened above 0.8℃were adopted. The rats were randomly divided into four groups namely the control group (model group), aspirin control group (aspirin group),Shujietuire Decoction with low-dose group (low-dose group),Shujietuiretang Decoction with high-dose group (high-dose group) and each group had 10 rats. Every group was dosed with method of lavage once. Normal and Model groups were dosed with distilled water 10ml/kg. The High-dose group was dosed with Shujietuire Decoction (2.1g/kg) and the Low-dose group was dosed with Shujietuire Decoction (1.1g/kg). The A.P.C group was dosed with aspirin compound (0.1g/kg). The rats were measured body temperature after dosed 1h, 2h, 3h, 4h respectively.(2)Effect on the contents of PGE2 and cAMP in hypothalamus tissue: The febrile rats were the same as the above experiment. Each group had 20 rats. Every group was dosed with method of lavage once. Normal and Model groups were dosed with distilled water 10ml/kg. The High-dose group was dosed with Shujietuire Decoction (2.1g/kg) and the Low-dose group was dosed with Shujietuire Decoction (1.1g/kg). The A.P.C group was dosed with aspirin compound (0.1g/kg). After dosed 2h, 4h and measured body temperature, the rats were decapitated and taken brain quickly in one minute. We took the hypothalamus tissue and measured the contents of PGE2 and cAMP, this step must go according to radioimmunoassay(RIA). Then we run the body temperature values of each group associated with the content analysis.Result: (1) Effect on body temperature of febrile rats induced by yeast: Eight hours after injected yeast, the body temperature of Model, High-dose, Low-dose and A.P.C groups increased obviously, which compared with Normal group(P<0.01). It illustrated that making febrile model was successful. After dosed, the body temperature of High-dose, Low-dose and A.P.C groups could fetch down. And compared with Model group, the difference of the temperature was significant (P<0.01). The body temperature of High-dose group was obviously lower than Low-dose and A.P.C groups after dosed (P<0.01).(2) Effect on the contents of PGE2 and cAMP in hypothalamus tissue: Compared with Model group,the difference in the contents of PGE2 and cAMP between High-dose, Low-dose, A.P.C groups was significant after dosed ( P < 0.01).The difference between High-dose and Low-dose,A.P.C was significance in statistic ( P < 0.01; P<0.05 ). Among them, high-dose group PGE2 cyclic adenosine monophosphate ( cAMP ) levels compared with control group was no statistically significant difference ( P>0.05 ),with low-dose, APC compared the differences were statistically significant (P<0.05). After the administration of 2h, 4h normal group, model group, high-dose group and the corresponding temperature values of PGE2,cAMP content of correlation analysis results show that there are obvious between a positive correlation (P<0.01).Conclusion: Our experiment proved that Shujietuire Decoction cound inhibit obviously the rise of body temperature of febrile rats induced by yeast. This showed it had antipyretic effect. Its mechanism of antipyretic effect may be that it may exert its antipyretic effect by reducing the content of neural febrile medium through some kind of means.
