Objective:External contraction fever of the childhood is one of the common disease in clinic, which has the features of falling urgently, spreading quickly and much cross-infection. It does much harm to the children. The disease frequently happens to upper respiratory tract infection, influenza and so on in western medicine. Meanwhile ninety percent of the upper respiratory tract infection is vivus infection. The western medicine mainly cure it by defervescing, antivirus and so on. Compared with them, the Chinese traditional medicine has many advantages in curing the disease, such as sustaining effect, few bounce-backs. Professor JiaoPing adopted ShuJieTueRe Decoction(SJTRT) to cure the childhood external contration about ten years, which has obtained better clinical curative effect.Professor JiaoPing regards that before the disease, the most children have the medical history ueberessen. It causes the disease because of long ueberessen, hot of the stomach intestines, external contraction and wind-irregular, and effect on inside and outside. So it often follows such as the difficult of constipation, the little yellow urinate, the red swelling of throat and so on. It is difficult to control the disease with one method. In order to get the best treating effect, we combine defervescing, relieving sore throat and detumescence and other ways.The patients get it easily and then it changes quickly. Once the out-evil get into the inside lf the body, the patients often have the hot of XueFen. The dug of cool the blood in creasing the contents of Shendi and Yuanshen is based on relieving Exterior Disorder and defervescing. It can help defervesce, moisten the intestines, clean waste of bodies out. So it can achieve the purpose of cleaning waste of bodies out, defervescing and not impairment of yin. The recipe has effect on getting rid of wind-heat and posion, defervescing, relieving sore throat and detumescence, nourishing yin and cool the blood and so on. It can help recover the normal body temperature better and quickly, and improve the clinical symptoms. It hasn′t toxic and side-effect. We chose febrile model of rats induced by yeast, observe its effect on defervescing, explore the function and mechanism of defervescing, and prove the recipe reasonable, in order to direct the practice of clinic furtherly.Method:The experiment needed healthy male Wistar rats that weighed about 180g to 200g. The process of the experiment was divided into two parts.(1) Effect on body temperature of febrile rats induced by yeast:Before the experiment, we made the rats adapt surroundings for three days in the laboratory of 18~22℃. They could move, eat and drink freely, and they measured body temperature twice every day. During the experiment, marked the rats first, and measured body temperature three times in the same way. The interval was half an hour. The rats whose fluctuations in temperature exceeded 0.3℃were rejected. Counting the average value of the temperature regarded as the basic body temperature. The rats were divided into five groups. Expcept for Normal group, the rest rats were injected 20% suspension yeast in the subcutis of back by 10ml/kg. After eight hours the rats whose body temperature heightened above 0.8℃were adopted. The rats were randomly divided into four groups:Model, High-dose, Low-dose and Aspirin compound(A.P.C), and each group had 10 rats. Every group was dosed with method of lavage once. Normal and Model groups were dosed with distilled water 10ml/kg. The High-dose group was dosed with SJTRT(2.1g/kg) and the Low-dose group was dosed with SJTRT(1.1g/kg). The A.P.C group was dosed with aspirin compound (0.1g/kg). The rats were measured body temperature after dosed 1h, 2h, 3h, 4h respectively.(2)Effect on the contents of prostaglandinE2(PGE2)and interleukinin-1β(IL-1β) in hypothalamus tissue and white blood cell(WBC) and neutuophil(NEUT) in blood of febile rats inducde by yeast: The febile rats was the same as the above experiment. The rats were randomly divided into four groups:Model, High-dose, Low-dose and Aspirin compound(A.P.C), and each group had 20 rats. Every group was dosed with method of lavage once. Normal and Model groups were dosed with distilled water 10ml/kg. The High-dose group was dosed with SJTRT(2.1g/kg) and the Low-dose group was dosed with SJTRT(1.1g/kg). The A.P.C group was dosed with aspirin compound (0.1g/kg). After dosed 2h, 4h, and measured body temperature, the rats were decapitated and taken brain quickly in one minute and taken blood quickly. We took the hypothalamus tissue and measured the contents of PGE2 and IL-1β, this step must go according to radioimmunoassay(RIA). Then we run correlation analysis of the tempature with the contents of PGE2 and IL-1βof every group. Then we checked the contents of WBC and NEUT in the blood, run correlation analysis of the tempature with the contents of WBC and NEUT of every group.Result:(1) Effect on body temperature of febrile rats induced by yeast:Eight hours after injected yeast, the body temperature of Model, High-dose, Low-dose and A.P.C groups increased obviously, which compared with Normal group(P<0.01). It illustrated that making febrile model was successful. After dosed, the body temperature of High-dose, Low-dose and A.P.C groups could fetch down. And compared with Model group, the difference of the temperature was significant (P<0.01). The body temperature of High-dose group was obviously lower than Low-dose and A.P.C groups after dosed 2h,3h,4h(P<0.01).(2) Effect on the contents of prostaglandinE2( PGE2)and interleukinin-1β(IL-1β) in hypothalamus tissue and white blood cell ( WBC) and neutuophil (NEUT) in blood of febile rats inducde by yeast: Compared with Model group, the difference in the contents of PGE2 and IL-1βbetween High-dose , Low– dose , A.P.C groups was significant after dosed 2h ( P<0.01 ) . The difference between High-dose and Low-dose , A.P.C were significance in statistic ( P < 0.01; P<0.05).The contents of PGE2 and IL-1βof every group decreased in some ways after dosed 4h than 2h. High-dose, Low-dose and A.P.C groups were greatly different with Model group ( P<0.01). There was no significance in statistic when the content of PGE2 in High-dose group was compared with Normal group(P>0.05). But there was significance when compared with Low-dose and A.P.C group(P<0.01). The content of IL-1βin High-dose group was compared with Low-dose and A.P.C group, the difference was significance in statistic ( P < 0.01). Run correlation analysis of the tempature of Normal, Model and High -dose groups with the contents of PGE2 and IL-1βafter dosed 2h and 4h. The result proved that there was positively correlated, the coefficient correlation was 0.859, 0.862, 0.770 and 0.727, P<0.01.After dosed 2h, there was significant difference between Model group and High-dose, Low-dose, A.P.C groups on the content of WBC and NEUT ( P<0.01). And compared with High-dose, the difference between Low-dose, A.P.C was significance in statistic(P<0.01,P<0.05).After dosed 4h, the average of the content of WBC and NEUT except Normal group decreased in some way. The content of WBC and NEUT of High-dose, Low-dose, A.P.C groups were greatly differences with Model group (P<0.01). There was no significance in statistic when compared High -dose group with Normal group(P>0.05). However, there was significance in statistic when the High-dose group was compared with Low-dose and A.P.C groups(P<0.01).Conclusion:(1)Our experiment proved that SJTRT had the antipyretic effect, and the efficacy of the drug was reliable. Its mechanism of antipyretic effect by reducing the release of central febrile medium through some kind of means.(2)The experiment proved that the recipe of SJTRT was reasonable, and explained the pathogenyof the fever induced by extrnal infection in childhood.(3)Through the experiment, we found that the change of the set point of thermotaxic centers was positively correlated with the content of the PGE2 and IL-1β.
