Background:In our protopased animal experiment, we had obsreved changes of mircocirculation post high-voltage electric burn , and demonstrated the existence of microcirculation disorder in high-voltage electric burned animal modles . It is an important phenominon of microvessel diversity that microvessel diastole contraction and endothelium injury . AngiotensinⅡcan make the vessel contract and advance secretion of NO . And acheved the accommodation of vessel . Solubility Vascular Cell Adhesion Molecule-1 can be the index of vessel injury. Accordingly , we did this experiment and observed changes of angiotensinⅡand Vascular Cell Adhesion Molecule-1 in blood serum , investigated the pathogenesis of microcirculation disturbance and treatment in early high_voltage electric injury stage.Objective:To observe changes of angiotensinⅡand Vascular Cell Adhesion Molecule-1 in blood serum and the effect of pentoxifylline (PTX) , and investigate the pathogenesis of microcirculation disturbance in early high_voltage electric injury stage.Methods:180 adult SD rats were randomly divided into electricity group , control group and treatment group . And the concentration of angiotensinⅡand Vascular Cell Adhesion Molecule-1 were measured by enzyme-linked immunosorbent assay(ELISA) at the T0(before),T1(instant) ,T2(1 hour after high_voltage electrical injury) , T3(2 hour after high_voltage electrical injury), T4(4 hour after high_voltage electrical injury) and T8(8 hour after high_voltage electrical injury) time phase . In each time phase ,there were 10 rats that were regardless of sex .And the female rats had not been pregnant.The body weight of SD rats'varied from 320g to 380g .Animal model was established , voltage at 10000V, electric current at 1.83±0.15A and electric burns time in 3 seconds . Blood serum of the rats collected was took suction from rats'heart .And each rat could offer volume of blood about 8ml-10ml. Specimen shoud be standing 30 minutes and centrifugating with 1000r/min about 15 minutes. Data is the measurement data .And SPSS 13.0 statistics software is also applied to analyze the data. Analysis of Variance of Factorial Design Data was applied to analyze main effect of different time and different handling factors and interaction effect .And one-way ANOVA was applied to analyze single effect . Further more, Student-Newman-Keuls(SNK-q) is also applied to mean intercomparison. Significance level is P<0.05 .Results :1 Results of Ang-Ⅱ1.1interaction effect results of Ang-Ⅱ1.1.1 The blood serum levels of Ang-Ⅱat different time took significantly changes (P<0.05). At the T0 and T1 time phase , T2 T3 T4 time phase , T3 T4 T5 time phase , there were non-significantly changes in the different groups(P >0.05) .1.1.2 The blood serum levels of Ang-Ⅱin different groups took significantly changes (P<0.05). Between electricity group and treatment groups there were non-significantly changes in the different groups(P >0.05) .1.1.3 There were interaction between handling factor and time .And , there were significantly changes .1.2 Single effect results of Ang-Ⅱ1.2.1 Different time results intercomparison in every group1.2.1.1 Control : Intercomparison is non-signification1.2.1.2 Electriction : There is non-signification between T0 and T1 .Among T3 T2 T5 T4 ,there is non-signification . The rest of group comparison is signification .Conclusion:1 The content of Ang-Ⅱand sVCAM-1 significantly increases in early high_voltage electric injury stage . It can be the index of microcirculation disturbance .2 The treatment of PTX can decrease the content of Ang-Ⅱ,and it decreases with the time .3 The changes of sVCAM-1 content can reflect the therapeutic effect of PTX .
