Atherosclerosis is considered to be a chronic inflammatory disease. Monocyte adhesion to endothelial cells is an important initial event at the onset of atherosclerosis.Smilax glabra Roxb belongs to heat-clearing and detoxifying drug,which has the effect of regulation of immunity and anti-inflammation. Precious studies have showed that Smilax glabra Roxb has the effect of anti-atherosclerosis. We try to find out the effect of Smilax glabra Roxb on adhesion of monocyte to endothelial cell and the expression of cellular adhesion molecules of HUVEC, so as to investigate the anti-atherosclerotic effect of Smilax glabra Roxb.This experiment consists of three parts:1. Effects of Smilax glabra Roxb on the adhesion of monocyte to IL-1 induced endothelial cells.OBJECTIVErTo observe the effects of SGR on monocyte adhesion to HUVEC and the toxicity effect on HUVEC. METHODS: (l)The dog was feeded with SGR so that the serum containing herbage was prepared. (2)The cellular viability were measured by MTT assay.(3) The adhesion of monocytes to HUVEC were measured by MTT assay. RESULTS: HUVEC was incubated with different concentrations of serum containing herbage and IL-1 for 12h. Different concentrations of Serum containing herbage of SGR ( 10% and 20%) obtained at lh, 2h and 6h markedly inhibite the monocyte-endothelial cell adhesion. Compared with 15% serum without herbage, the cellular viability of HUVEC indubated with 15% serum containing herbage obtained at 75min increase obviously. CONCLUSIONS: Serum containing herbage of SGR obtained at llu 2h and 6h have the effect of inhibiting the monocyte-endothelial adhesion. Serum containing herbage SGR has unconspicuous damage on HUVEC.2. Effect of SGR on the expression of ICAM-1 of HUVEC induced by IL-1OBJECTIVE: To examine the effect of SGR on the expression of ICAM-1 of HUVEC induced with IL-1. METHODS: The expression of ICAM-1 was measured by Flow Cytometry. RESULTS: 20% serum containing herbage obtained at 45min decreased the expression of ICAM-1(P <0.05); 10% and 20% serum containing herbage obtained at 60min decreased the expression of ICAM-1 markedly(P<0.01). CONCLUSIONS: There may be some ingredients in serum containing herbage of SGR that inhibite the expression of ICAM-1.3. Effect of SGR on the expression of VCAM-1 of HUVEC induced by IL-1OBJECTIVE: To examine the effect of SGR on the expression of VCAM-1 of HUVEC induced with IL-1. METHODS: The expression of VCAM-1 was measured with Flow Cytometry. RESULTS: 5% and 10% serum containing herbage obtained at 6h and 12h decreased the expression of VCAM-1 markedly (P<0.01). CONCLUSIONS: There may be some ingredients in SGR containing serum inhibit the expression of VCAM-1.On the whole, serum containing herbage of SGR has the inhibition effect of expression of ICAM-1 and VCAM-1, then inhibit the adhesion of monocyte and endothelial cell. This may be involved in the machanism of anti-arthrosclerosis of SGR herbage.
