Abstract:Objective:To study on effect and mechanism of Donepezil in treating the Alzheimer's disease in rat, to observe the application of the instrument of tri-cylinder intercommunication in the animal ethologic changes. Methods:Thirty rats were randomly divided equally into the control group,AD group,Donepezil+AD group(Donepezil group). Aβ1-40 (2μg/μl)5μl was infused in the hippocamp of right for AD model by stereotaxis instrument. The rats of Donepezil group were Iavaged 2ml of Donepezil(6.79×10-2mg/ml) everyday for twenty eight days at infusing Aβ1-40 seventh days. The rats of control and AD group were lavaged 2ml of di-parboil water everyday for twenty eight days. Then, the changes in time for searching the eclipse period of platform were measured in Morris water maze, sametime, the changes in the distance for searching food were measured in the instrument of the tri-cylinder intercommunication. The luidine blue 5ul was infused in the hippocamp of right for locating of the Aβ1-40 by the stereotaxis instrument. The changes in tissue of the right hippocamp were observed by HE of dyeing in microscopy. The activity of SOD and content of MDA were determined right and left of the hippocamp. The experimental data was expressed as mean±standard deviation (x±s), and deal with the SPSS13.0 software of statistics, p<0.05 for statistical significance. Results:1. The toluidine blue was distributed in injection site of Aβ1-4o for showing AD successful mode.2. The changes in the structure of hippocamp:The pyramidal cell and nerve fibre were round and order, cell nucleus were round, nucleolus were clear in control group. pyramidal cell and nerve fibre were decrease and disorder in AD group. However, pyramidal cell and nerve fibre were round and order in Donepezil group.3. The changes in the searching the eclipse period of platform:The changes in the searching the eclipse period of platform in AD group (28.64±25.44) compared with the control(13.85±8.40)and Donepezil group (12.99±4.40), there were the significant differences (p<0.05).4. The change in distance for searching food:The change in distance for searching of food in AD group (16.65±12.77) compared with the control (7.57±4.82) and Donepezil group(9.92±8.05), there were the significance differences. 5.The change in the activity of SOD and content of MDA:The change in the activity of SOD in AD group (125.36±6.57) compared with the control (146.69±5.35) and Donepezil group(133.68±3.45), there were the significant differences. The change in the content of MDA in AD group (49.48±1.73) compare the control (28.53±1.21) and Donepezil group(34.41±1.29) there were significant differences. Conclusion:1.The mode of AD was successful.2. AD could be resuled by in hippocamp.3. The pathologic changes in hippocamp were reduced with Donepezil for removing of the free radicle.4. The tri-cylinder intercommunicate could be used for studying of animal behavior.
