| Alzheimer’s disease(AD)is a kind of primary nervous system degenerative diseasehappened in old age or presenium.Patients with this disorder have sustained higher nervous function,and the reduction of memory,sensory ability,judgment ability,thinking ability,the analysis judgment ability,visual space recognition ability,athletic ability without cognitive deficits.With the development of modern medical science,the acceleration of social aging process,Alzheimer’s disease has become one of the diseases which may have serious threat to human life and health.Donepezil,the Cholinesterase inhibitor,increases the concentration of acetylcholine in synaptic cleft,so as to resume the function of cholinergic nerve,as the result,the symptoms of alzheimer’s patients obviously alleviated,the quality of the patients’ life improved.Therefore,Donepezil is widely used in the treatment of Alzheimer’s disease.Chrysophanol is extracted from RheumofficinaleBaill.,a Traditional Chinese medicine.Studys found that Chrysophanol can improve symptoms of Alzheimer.In recent years,the method,which combines traditional Chinese with western medicine to treat Alzheimer’s disease,has been reported in literatures and obtainedsatisfactory curative effect.This topic used the method of combining traditional Chinese with western medicine,that is,Donepezil and the Chrysophanol,on curing Alzheimer’s disease model rats treated,in order to obtain satisfactory curative effect.Objective:To produce rat model of Alzheimer’s disease,and observe the effect of chrysophanol and Donepezil on the ability of learning and memory and the activity of ChE,ChAT,POD and T-AOC in the AD rat’s brain,and then discuss the preventive and treatment effect of chrysophanol and Donepezi on AD rat.The effects of Chrysophanol jointDonepezil onhymus and spleen indexof AD rat were detected,and the pathology of the brain of each rat were examined.Method:The AD model was produced by injecting D-galactose and AlCl3 intraperitoneally for90 days.Rats were randomly divided into normal group,model group,chrysophanolgroup(0.014mg/kg/d),Donepe-zilgroup(0.33 mg/kg/d),chrysophanoljoint Donepezilgroups(chrys-ophanol0.014mg/ kg/d+Donepezil 0.33 mg/kg/d,chrysoph-anol0.007 mg/kg/d + Donepezil0.66 mg/kg/d,chrysophanol0.028 mg/kg/d + Donepezil0.17 mg/kg/d).From the 20 th day of D-galactose and AlCl3 intraperitoneal injection,chrysophanol and Donepezil were intragastricadministrated respectively for 70 days.The learning and memory ability of each group rats were observed by Morris water maze.The activity of ChE,ChAT,POD and T-AOC in the AD rat’s brain were determinated.Result: RatsintragastricadministrationwithChrysophanol,Donepezil and chrysophanol joint Donepezilhad shorter latency(P<0.05,P<0.01)and more numbers through the platform(P<0.05,P<0.01).Compared with chrysophanol group and Donepezil group,rats in chrysophanol joint Donepezil group(chrysophanol0.028 mg/kg/d + Donepezil0.17 mg/kg/d)had shorter latency(P<0.05)and more numbersthrough the platform(P<0.05).Compared with the model group,the activity of ChAT,POD and T-AOC in the Chrysophanol group,Donepezil group and chrysophanol joint Donepezil groups rat’s brain elevated significantly(P<0.05),the activity of ChAT and T-AOC of the chrysophanol joint Donepezil group(chrysophanol0.028 mg/kg/d + Donepezil0.17 mg/kg/d)levatedsignificantly(P<0.01).The activity of ChE reduced(P<0.05).The thymus and spleen indexs of each dosage groupwere significantly higher than that of model group.There are some improves of the histopathological injury of the AD rats,but the improvement have no significance.While the improves of theChrysophanol jointDonepezil groups weresignificant.Conclusion: Chrysophanol jointDonepezil can improve the learning and memory ability of AD rats,their effects were collaborative.The effect is associated with their doses.Chrysophanolas the main drug,supplemented by Donepezil may get a better effect on Alzheimer’s disease. |
