The Investigation Of Quercetin Induce Apoptosis Of Prostate Cancer Cell PC-3

Objective:At present there is no effective treatment on prostate cancer especially hormonal independent prostate cancer, and people discover the advantage of cancer treatment by traditional Chinese medicine gradually. Nowadays, many research have found that quercetin may induce variety of tumor cells to apoptosis. We use quercetin to treat prostate cancer cell PC-3 which in vitro culture in this Research. We observe the condition of apoptosis, and we analysis results too.So we can make foundation for further empirical study and clinical work.Methods:First, we made an overview on the research of traditional Chinese medicine on prostate cancer. Then we made an overview on the hormonal independent prostate cancer's treatment of traditional Chinese medicine and the western medicine. We find that the cancer treatment of traditional Chinese medicine have great effect. Secondly, we made a description on antineoplastic mechanism of quercetin. Next, we begin with the empirical study. We culture human prostate cancer cell PC-3 in vitro first, and we begin with empirical study after we collect cells. We add quercetin in cells with different concentration. Then we detect effective concentration and test it with MTT assay, and we observe morphologic change by inverted microscope. We add quercetin in cells with different concentration again and observe the condition of cell apoptosis by flow cytometry. We observe the change of fine structure through transmission electron microscope after we add quercetin in cells with different concentration.Results:The results of MTT assay show that quercetin inhibited the growth of human prostate cancer cell PC-3,and the effect regulated both time-dependently and dose-dependently. The pictures of inverted microscope show that cells apoptosis. The results of flow cytometry show that quercetin can induce apoptosis of it, and the effect regulated both time-dependently and dose-dependently. Pictures of transmission electron microscope show that cells apoptosis.Conclusion:Quercetin inhibited the growth of human prostate cancer cell PC-3 which in vitro culture, and quercetin can induce apoptosis of it. The effect regulated both time-dependently and dose-dependently.
