With these results, we wanted to put our discoveries into the clinicopathological diagnosis, prognosis evaluation and gene therapy.Material and Method We used SP immunohistochemistry method to detect the expressions of Caspase-3,Fas and FasL in tissue microarray with 270 cores, and analyzed the relationship between their expressions and clinicopathological parameter.Results 1. In normal lung cells,Fas is express in cell membrane;Fas-positive tumors demonstrated a diffuse cytoplasmic staining without cell surface expression.The positive rate of Fas in primary lung cancer,lymph node metastasis lung cancer and precancerous lesion were lower than that of normal lung tissue(P<0.05). The positive rate of Fas was significantly lower in SCLC than NSCLC(P<0.05).2. The positive rate of FasL in primary lung cancer,lymph node metastasis lung cancer and precancerous lesion were higher than that of normal lung tissue(P<0.05). The positive rate of FasL was higher in the group ofâ…¢+â…£stage thanâ… +â…¡stage (P<0.05), and it was higher in the group with lymph node metastasis than that without lymph node metastasis (P<0.05).3.The positive rate of Caspase-3 in primary lung cancer,lymph node metastasis lung cancer and precancerous lesion were lower than that of normal lung tissue(P<0.05). The expression of Caspase-3 was higher in the group of high-middle differentiation than low-undifferentiation, The positive rate of Caspase-3 was higher in the group ofâ… +â…¡stage thanâ…¢+â…£stage, higher in the group without lymph node metastasis than with lymph node metastasis(P<0.05).4. There was significantly negative correlation between Fas and FasL(P<0.05).Conclusions1. The positive location of Fas is cell membrane in normal lung cells;but is plasma in lung caner cells,which indicate that ectopic expression of Fas is related with immune escape of lung caner cells. 2. The positive rate of Fas was lower in primary lung cancer,lymph node metastasis lung cancer and precancerous lesion than that in normal lung tissue, which suggested that Fas promote the genesis and progress of lung cancer. Fas may have important value in early diagnosis of lung cancer.3. The positive rate of Fas was significantly lower in SCLC than that in NSCLC, which suggested it can be a marker to distinguish SCLC and NSCLC.4. The positive rate of FasL was higher in primary lung cancer,lymph node metastasis lung cancer and precancerous lesion than that in normal lung tissue, which suggested that FasL promote the genesis and progress of lung cancer. FasL may be a new index for genesis forecast of lung cancer.5. The positive rate of FasL was significantly related to the degree of clinical stages and lymph node metastasis, which suggested it can promote the malignant transformation and progression, so it has a reference in prognosis evaluation.6. The positive rate of Caspase-3 was lower in primary lung cancer,lymph node metastasis lung cancer and precancerous lesion than that in normal lung tissue, which suggested that Caspase-3 promote the genesis and progress of lung cancer. Caspase-3 may have important value in early diagnosis of lung cancer.7. The positive rate of Caspase-3 protein was higher in the group of high-middle differentiation than low-undifferentiation, higher in the group ofâ… +â…¡stage thanâ…¢+â…£stage(P<0.05), higher in the group without lymph node metastasis than with lymph node metastasis(P<0.05).These suttested that Caspase-3 promote the progress of lung cancer,and it can be a reference in prognosis evaluation.8. There was significantly negative correlation between Fas and FasL,which indicate that the express of Fas depress and FasL increase in primary lung cancer promote the genesis of lung cancer.9. Tissue microarray has the advantage of high throughput, high efficiency, saving reagent and time.
