Objective:Gastric adenocarcinoma is one of the most common gastrointestinal tumors in our country, which has a high mortality. It is very significance to investigate the pathogenesis and progression of gastric cancer. The tumorigenesis of gastric cancer is a serially dynamic process concerned of the activation of many oncogenes, inactivation of anti-oncogenes and the subsequent changes of cytobiological behaviors. Lots of oncogenes and anti-oncogenes play different roles in different biochemical pathways. They interact each other to regulate the occurrence and development of tumor. As other tumors, the mechanism of gastric carcinogensis has not been clear. Recent studies showed that CDX2,COX-2 and NF-κB play important roles in occurrence, development and invasion of gastric cancer.This study detected the expression of CDX2,COX-2 and NF-κB in the tissue of gastric cancer, precancerous lesions and normal gastric mucosa, and to discuss the correlation of CDX2,COX-2 and NF-κB expression,and to guide the early dignosis or judge prognosis of gastric carcinoma and provide theoretical foundation for prevention and clinical treatment.Methods:The formalin-fixed and paraffin-embedded tissues from 60 cases of gastric carcinoma,45 patients of precancerous lesions and 20 cases of normal tissues were evaluated for the expression of CDX2, COX-2 and NF-κB by SP and SABC immunohistochemistry, the levels of these proteins in tissues with different types and their correlation with gastric carcinoma were analyzed. And the biological behavior of gastric carcinoma were analyzed,including patients' sex, age, size, differentiation degree, infiltration depth and lymph nodes metastasis.All statistical analysis were carried out with SPSS 16.0, and P<0.05 was considered significant.Results:There was a significantly much higher positive rate of NF-κB and COX-2 proteins in gastric carcinoma than that in precancerous lesions group and normal tissues group (P< 0.05 respectively). CDX2 was expressed in gastric carcinoma group and precancerous lesions group, while low expressed in the normal mucosa.Positive rates of CDX2 expression in precancerous lesions group was higher than gastric carcinoma group (P<0.01). CDX-2 was closely correlated with differentiation degree, gross tumor volume and infiltration. COX-2 was correlated with gross tumor volume, invasive depth and lymph node metastasis. NF-κB showed positive expression of differentiation degree, gross tumor volume, invasive depth and lymph node metastasis. NF-κB expression was positive correlated with COX-2 in gastric carcinoma(P<0.01),while CDX2 expression was negative correlated with COX-2 and NF-κB in gastric carcinoma(P<0.01).Conclusion:1. The positive rates of COX-2 and NF-κB gradually increased with the progression of gastric mucosa lesions, respectively. Expression of protein CDX2 in precancerous lesions tissues was significantly higher than gastric carcinoma tissues and normal tissues. It shows that CDX2, COX-2 and NF-κB were in administrative relation and cooperated in the process of gastric carcinoma development,which might promote gastric carcinogenesis as some stimulating factors in the early events. It is suggested that the changed expression of CDX2, COX-2 and NF-κB might be of value in guiding the clinical early dignosis, the prevention and treatment.2. The overexpression of COX-2 and NF-κB were correlated with infiltration,gross tumor volume and lymph nodes metastasis. And the expression of CDX2 were correlated with differentiation degree,infiltration and gross tumor volume.It was indicated that the expression of them closely correlation with the development and invasion of gastric carcinoma.It is suggested that the positive expression of CDX2,COX-2 and NF-κB might be of value in judging the prognosis of gastric carcinoma.3. NF-κB expression was positive correlated with COX-2 in gastric carcinoma and may have synergistic effect on the carcinogenesis and development of gastric carcinoma; CDX2 expression was negative correlated with COX-2 and NF-κB in gastric carcinoma.They may contribute to the development and progression of gastric carcinoma incoordination.
