The Expressions Of C-kit, Cyclin D3 In The Spermatogonia Undergo Body Cavity Temperature

Objective:To study the relationship that the expression of c-kit,Cyclin D3 mRNA and protein in type A spermatogonia of the different periods experimental cryptorchidism,in order to provide a basis research about mechanism for temperature induced dyszoospermia in spermatogonia stage.Methods:After establishing cryptor-chidism model of adult SD rat animal,the type A spermatogonias were selected out respectively from testis at the time on 7th days,14th days,21th days,which can be achieved by Speed centrifugation and difference attachment.The expressions of c-kit,Cyclin D3 mRNA and protein were detected by RT-PCR and Western Blot,Meanwhile,the FCM was used to detect the type A spermatogonias cell cycle.Immunohistochemistry c-kit to detect the distribution of type A spermatogonia of testis at term.Results:The expression mRNA and protein of c-kit and Cyclin D3 were detected in the type A spermatogonia in normal SD rats.lt was significantly up on the 7th day in the cryptorchid rats,onl4th days decreased and to be a minimum on 21th days,but still higher than the control group The expression of c-kit,Cyclin D3 mRNA and protein was consistent with the trend. And expression of c-kit and Cyclin D3 was statistically significant between each experimental group with the normal group(P<0.05).For 7th days group,21th day and 14th days group,the comparison between two groups each other was statistically significant (P<0.05).FCM analysis: the cell cycle of experi-mental group and control group was no difference cell cycle.Conclusion:The arrest of cryptorchidism Spermatogonial cells may not be related to the expression of c-kit of spermatogonia,namely,the body cavity temperat-ure may not be through the SCF/c-kit and the downstream signaling to Cyclin D3 system blocked spermatoge-nesis spermatogonia.
