Objective To study the role and expression of miRNA-101(miR-101)and Ran-binding protein 9(Ran BP9)in cryptorchidism testes.Methods Sixty 6-week-old male ICR mice were randomly divided into two groups.Mice in cryptorchidism group(Cry group)underwent operation to fix left testes with fat pad onto peritoneum.Mice in the control group(Con group)underwent sham operation that the left testes were kept in the scrotum.Right testes of mice in Cry group were used as self-controls(SC group).The mice were sacrificed at 3,7,14,21 and 28 days after operation.The body weight and testicular weight of the mice were recorded.The histological changes were examined by HE staining.Western blot,q RT-PCR and immunofluorescence were used to detect the expression of miR-101 and Ran BP9.Results The testis index of mice in cryptorchidism group was significantly decreased at 7 days after operation compared with those in SC group and Con group(P<0.05).HE staining showed impaired testis structure in cryptorchid mice at 7 days after surgery.The expression of miR-101 was significantly increased in the cryptorchidism group while the expression of Ran BP9 was decreased(P<0.05).Conclusion The expression of miR-101 was increased in cryptorchidism while Ran BP9 was decreased in cryptorchidism,suggesting that miR-101 and Ran BP9 may contribute to spermatogenesis disorder in cryptorchidism.
