Object:To construct the mode of allogeneic skin transplantation experiment on rabbits, and observe the general changes, skin graft beginning rejection time and survival time of rabbits and the changes of T Lymphocyte subsets before and after the injection of recombination human interleukin-10, to explore the mechanism of recombination human interleukin-10 in transplantation of the rabbits skin..Methods:1.allogenetic skin transplantation experiment on rabbits:There were six groups in receptor groups, three rabbits in every group; eighteen in donor groups. Had the hair cut on rabbits back before graft, removed two skin transplants of 1.5cm×1.5cm sizes each donor and receptor rabbit, single cage raised. Intramuscular administration 1 day before the graft trials and totally 10 days, observed grafted skin and diet situation in each group. 2.Flow Cytometer:detected the changes of maternal serum CD3+,CD3+CD4+ CD3+CD8+,CD3+CD4+/CD3+CD8+ level in six groups 1 day before graft and 1 day,4 days,7 days,14 days,21 days,28 days after graft with direct double-colour fluorescence labeling method.Result:1. general condition:the rabbits of high CsA group had a poor diet and mental state and hair loss phenonmenon; skin condition:dry dressing, soft and red skin, medium tension, and no leakage around the tidy 3 days after graft.2. skin graft beginning rejection time:Low doses of hIL-10 group6.67±2.08 d; high doses of hIL-10 group 9.00±3.60 d; low doses of CsA group 12.00±1.00 d; high doses of CsA group 19.67±1.15 d; combined group 16.67±1.15 d; negative control group 4.33±0.85 d. In addition to low doses of hIL-10, each group and control group had a significant difference, p<0.05.3.survivial time of the skin:Low doses of hIL-10 group 13.33±3.05 d; high doses of hIL-10 group 16±2.0 d; low doses of CsA group 15.67±0.57 d; high doses of CsA group 21.33±2.52 d; combined group 19±1.73 d; negative control group 7.67±1.15 d. every experiment group and negative control group had a significant difference, p<0.05.4. In the event of acute rejection, CD3+CD4+ T lymphocyte and CD3+CD4+/CD3+CD8+ level increased, while CD3+CD8+ level and the incidence of rejection was no significant correlation.Conclusion:1. Animal experiments showed that the recombination hIL-10 inhibited the rejection on allogeneic skin graft, delayed the time of skin graft rejection, extended the survival time of grafted skin, and had a synergistic effect with CsA.2. recombination hIL-10 inhibited CD3+CD4+ T lymphocyte activation, reduced the IL-2 and IFN-y production, induced the conversion Thl to Th2, delayed the occurring of acute rejection by inhibiting the activation of CD3+CD4+ lymphocyte.3. There was a certain correlation among CD3+CD4+/CD3+CD8+level and the beginning rejection time and the survival time of the grated skin, the ratio had rose before the grafted skin rejected, and had reached the peak when they necrosed. It indicated that the ratio can be monitored the rejection. CD3+ level elevated after transplantation because of the activation and proliferation of CD3+ T lymphocyte stimulated by the foreign antigen.
