遵义医学院硕士研究生毕业论文Objective:Investigating the effect of Dendrobium nobile (D. nobile) to advanced glycation end products (AGEs) and its receptor gene expression in diabetic rat nephrogenic tissue, we preliminary study the protective mechanism of D. nobile. to diabetic nephropathy in rats.Methods:The thirty rats were divided into three groups randomly, each group included ten rats. They were control group, model group and D. nobile preventive group. Except control group, the other groups were injected once with streptozotocin (55mg/kg body weight) intraperitoneally. Then the rats in D. nobile preventive group were administrated D. nobile 10g/kg for 12 weeks intragastrically. The levels of blood glucose were assayed by the glucose meter. The kidney function was examined by the blood analyzer. The histopathology of kidney and pancreas were examined by HE staining. The AGEs in serum were detected by using fluorescence spectrometry. The levels of RAGE protein in rat nephrotic tissue were detected by immunohistochemical technique semiquantitativly. Real-time fluorescence quantitative PCR was used for detection of the level of expression of RAGE mRAN.Results:(1) The diabetic rat model was established successfully. The rats in model group showed increased glomerular volume nodular glomerulosclerosis, hyalinosis, mesangial thickening, but neither in control group nor D. nobile preventive group. (2) After 12 weeks, three groups of rats in body weight, kidney weight/body weight of the rats, the levels of blood glucose, urea nitrogen, serum creatinine, urine creatinine,24-hour urine protein and serum AGEs were statistically significant (p<0.05). (3) There was a significant difference in the levels of RAGE protein in each group (p<0.05). D. nobile preventive group compared with the model group, the RAGE protein significantly reduced. (4) Real-time fluorescence quantitative PCR results confirmed that the control group, model group, D. nobile preventive group were statistically significant in expression levels of RAGE mRNA (p<0.05), the level of expression of RAGE mRAN in D. nobile preventive group was significantly lower than in model group. Conclusion:(1) D. nobile can reduce the levels of blood glucose of diabetic rats. (2) D. nobile can reduce the renal pathological damage in diabetic rats, decreas the levels of AGEs in serum, reduce the levels of RAGE protein and RAGE gene expression in diabetic rats kidney tissue.
