Objectives:The purpose of the study was to detect the expressions of Stromal Cell-Derived Factor-1(SDF-1) and its receptor CXCR4 in gastric adenocarcinoma, to evaluate their roles in carcinogenesis, development, infiltration and metastasis as well as their clinical significances, to provide new axiliarily diagnostic marker for early finding and new target spots for targeted therapy.Methods:1.The expressions of SDF-1 and its receptor CXCR4 were detected by SP immunohistochemical method in 60 cases of gastric adenocarcinoma and 30 cases of normal gastric mucosa.2.The expressions of SDF-1mRNA and CXCR4mRNA were detected by RT-PCR method in 29 cases of gastric adenocarcinoma,15 cases of normal gastric mucosa.Results:1.the results of immunohistochemistry The differences of SDF-1 and CXCR4 proteins positive expression among the gastric adenocarcinoma and the normal gastric mucosa were significant (P<0.01, P<0.01); The positive expressions of SDF-1 and CXCR4 in gastric adenocarcinoma groups both were higher than those in the normal gastric mucosa groups. The positive expressions of SDF-1 and CXCR4 were both closely correlated with the metastasis of lymph node:the positive expressions of SDF-1 and CXCR4 in gastric adneocarcinoma with lymphnode metastasis were obviously higher than those without lymphnode metastasis (P<0.01, P<0.05); But both of them were not related to gender, age of patients, differentiation of carcinoma and the depth of tumor infiltration (P>0.05).2. the results of RT-PCR The expressional quantities of SDF-1mRNA, CXCR4mRNA in gastric adenocarcinoma both were obviously higher than those in the normal gastric mucosa (P<0.01, P<0.01). The expressional quantities of SDF-1mRNA and CXCR4 both were closely correlated with metastasis of lymph node: the expressional quantities of SDF-1mRNA and CXCR4mRNA in gastric adenocar-cinoma with lymphnode metastasis were obviously higher than those without lymphnode metastasis (P<0.01, P<0.01). But both of them were not related to gender, age of patients, differentiation of cancer and the depth of tumor infiltration (P>0.05).3.Correlation analysis (1)The positive expressions of SDF-1 and CXCR4 proteins are positively correlated in the 60 cases of gastric adenocarcinoma (r=0.310, P<0.05). (2) The expressional quantity of SDF-1mRNA and CXCR4mRNA are highly positively correlated in the 29 cases of gastric adenocarcinoma (r=0.875, P<0.01). (3) The positive expression of SDF-1 protein and expressional quantity of SDF-1mRNA are positively correlated in the 29 cases of gastric adenocarcinoma (r=0.649, P<0.01). (4) The positive expression of CXCR4 protein and expressional quantity of CXCR4mRNA are positively correlated in the 30 cases of gastric adenocarcinoma (r=0.556, P<0.01).Conlusion:1. SDF-1 and its receptor CXCR4 may play an important role in carcinogenesis progression and metastasis of gastric adenocarcinoma.2. To detect the expressions of SDF-1 and CXCR4 may provide guidance for the early finding, predicting prognosis of gastric adenocarcinoma.3. Further researches are needed to determine whether SDF-1 or(/and) CXCR4 can serve as target spots in the targeted therapy of gastric adenocarcinoma.
