Study On Effective Fractions From Gentiana Veitchiorum Hemel In The Treat To Disease Of Respiratory System

Objective:To investigate what is the active component of alcoholic extract of Gentiana veitchiorum Hemel to treat chronic bronchitis model in mice and hepatic fibrosis model in cells.Methods:Gentiana veitchiorum Hemel was extracted with alcohol. The alcoholic extract was dissolved in distilled water, and then extracted with ligarine,chloroform,ethyl acetate,n-butyl alcohol in order. Five groups were obtained. The chronic bronchitis model in mice was made by mice breathing ammonia water. 80 Mice were divided into 10 groups at random,except the control group and model group, other groups were intervened with corresponding drugs, and then all groups were detected the activity of SOD,T-AOC,TNF-α,IL-10 and pathological changes of each group were compared . Took lung tissue from 2 days newborn SD rats for the primary culture; Pulmonary fibrosis model was induced by factor-beta1(TGF-β1); Observed the effect of respective medicine by means of MTT assay and collagen determination. Results:Compared with the model group, the activity of SOD ,T-AOC the drug-intervening groups increased obviously , and the contents of TNF-α,IL-10 decreased significantly. Pathological changes of Lung tissue were compared showed alleviation of injury from chronic bronchitis in ethyl acetate group and n-butyl alcohol group. In the pulmonary fibrosis model ,ethyl acetate and n-butyl alcohol groups had obvious inhibition on the growth of collagen induced by TGF-β1(P<0.05 or P<0.01),In the MTT assay, compared with the results of the model group,the OD value of the water-course, ethyl acetate and n-butyl alcohol groups were significantly decreased(P<0.05 or P<0.01).The results indicated that ethyl acetate and n-butyl alcohol groups in the extract of Gentiana veitchiorum Hemel could significantly remiss pulmonary fibrosis.Conclusion:Ethyl acetate and n-butyl alcohol-extract had amelioration on chronic bronchitis and pulmonary fibrosis model. It's true that the active component of Gentiana veitchiorum Hemel for treating chronic bronchitis and pulmonary fibrosis were in them.
